盐穗木根际土壤中耐盐菌B6的分离与鉴定

         

摘要

[Objective] The analysis of the morphological, physiological and biochemical characteristics, 16S rDNA identification were conducted on the halotolerant bacteria B6 which was isolated from rhizosphere microbe of Halostachys caspica in Wujiaqu Xinjiang Province. [Method] The common primers for the 16S rDNA of eubacteria were used for polymerase chain reaction(PCR). Homology analysis was made by searching in Genbank with NCBI-Blast and the phylogenetic tree was constructed with MEGA4. [Result] Strain B6 can grow on the media containing 0 -20% NaCl, and was bacilli with spore forming. The amplified 16S rDNA fragment length was 1 446 bp and the result indicated that the 16S rDNA homologies were 99% between strain B6 and many kinds of Bacillus Licheniformises. [Conclusion] The tree showed the closest relationship was between strain B6 and Bacillus licheniformis. By analyzing the physio-biochemical characteristics of strain B6, strain B6 could be identified as a species of Bacillus licheniformis.%[目的]从新疆五家渠市耐盐碱植物盐穗木根际土壤中分离出兼性耐盐菌B6,并对其分别进行形态特征、生理生化特性分析及16S rDNA鉴定.[方法]采用真细菌16S rDNA通用引物PCR扩增B6菌株的16Sr DNA,使用NCBI-Blast软件在GenBank数据库中对其进行同源性检索,使用MEGA4软件构建系统发育树.[结果]B6菌能在0~20% NaCl的条件下生长,为有芽孢的杆状菌;B6的16S rDNA长度为1446bp,与多种地衣芽孢杆菌的16S rDNA序列同源性高达99%.[结论]系统发育树表明B6菌株与地衣芽孢杆菌的亲缘关系最近,结合生理生化分析,可判断B6菌株为地衣芽孢杆菌.
