紫杉醇脂质体对乳腺癌及卵巢癌细胞生长抑制作用的基础研究

         

摘要

Objective To analyze and research the inhibition effect of paclitaxel liposome for cell growth of breast cancer and ovarian cancer.Methods The Ovarian cancer SKOV-3 cell line and breast cancer MCF-7 cell line were collected as the research object, and the SKOV-3 cells and MCF-7 cells were treated with different doses of paclitaxel liposome.Analyzed the time-concentration relationship of tumor cells by liposomal paclitaxel inhibition based on cyto-logical level.Results With the same planting density, the cell growth of MCF-7 cells was faster then SKOv-3 cells. After the analysis on the time and concentration of paclitaxel liposome, we found that paclitaxel liposome has significant dose-time dependent effect on SKOV-3 and MCF-7 cells.With the prolonging of administration time, the inhibition effect of paclitaxel liposome with the same concentration on cells was increased.With the increasing of the concentration of paclitaxel liposome, the inhibition effect of paclitaxel liposome with the acting time on cells was increased.SKOV-3 and MCF-7 cells might be blocked in G2/M phase by paclitaxel liposome and paclitaxel.Conclusions Witht the lipo-some as the transporter, the incubation cycle and inhibition effect of paclitaxel on SKOV-3 and MCF-7 cells was not changed, so paclitaxel has good prospects for its clinical application.%目的:对紫杉醇脂质体对乳腺癌及卵巢癌细胞生长抑制作用进行分析和研究。方法将卵巢癌SKOV-3细胞系以及乳腺癌MCF-7细胞系作为研究对象,选择不同剂量的紫杉醇脂质体分别处理SKOV-3以及MCF-7细胞,然后立足于细胞学水平,对肿瘤细胞受到的紫杉醇脂质体的抑制作用的时间浓度关系进行分析。结果在具有相同种植密度的情况下,相对于SKOv-3细胞而言,MCF-7细胞较快的增长速度。在经过对紫杉醇脂质体作用时间和浓度进行筛选之后,可以发现其对SKOV-3以及MCF-7细胞的剂量-时间依赖效应非常显著。随着不断延长的用药时间,浓度相等的紫杉醇脂质体对细胞的抑制力也在不断的增加。随着药物浓度的不断增加,在作用时间相同的情况下紫杉醇脂质体对细胞的抑制力也在不断的增加。 SKOV-3以及MCF-7细胞会被紫杉醇脂质体以及紫杉醇阻滞在G2/M期。结论以脂质体作为转运载体之后,紫杉醇对SKOV-3以及MCF-7细胞的作用周期和抑制作用都没有得到改变,因此其临床应用前景良好。
