电针对WKY大鼠抑郁样行为及海马GluR1的影响

         

摘要

目的 通过电针“百会”“印堂”观察对抑郁大鼠行为及海马GluR1的影响.方法 21只雄性WKY大鼠被随机分为电针组、假针组和模型组;雄性Wistar大鼠7只分为正常组.电针组取穴为“百会”“印堂”,假针组给予安慰治疗,模型组和正常组常规饲养,21 d后测定各组大鼠糖水消耗量(SPT)、强迫游泳实验(FST)的改变,用Westem blot检测大鼠海马谷氨酸受体(glutamate receptorl,GluRl)蛋白表达.结果 电针组蔗糖水喜好率明显高于假针组(P<0.05),正常组蔗糖水喜好率高于模型组(P<0.05);强迫游泳试验中电针组和正常组不动时间少于假针组和模型组(P<0.05);海马GluR1的表达量中,电针组高于假针组(P<0.05),正常组高于模型组(P< 0.01).结论电针对WKY抑郁大鼠的行为学核心症状有明显的改善作用,其作用可能与海马GluR1的表达增加有关.%Objective To observe the effects of electroacupuncture at "Baihui" and "yintang" on depressive behavior and hippocampal GluR1 of WKY rats.Methods 21 male WKY rats were randomly divided into EA group,sham EA group and model group;7 male Wistar rats served as normal group.EA group select "Baihui'and "yintang",Sham EA group were treated with placebo.Model group and normal group were fed regularly.Sucrose preference test(SPT) and forced swimming test(FST) were measured in rats after 21 d.Protein expression of glutamate receptors in rat hippocampus was detected by Western blot.Results After treatment,sucrose preference rate was significantly higher in EA group than sham EA group (P<0.05),sucrose preference rate was significantly higher in normal group than model group(P<0.05);In forced swimming test,the immobility time in the EA group and the normal group was less than sham EA group and model group (P<0.05);The expression level of GluR1 in hippocampus was higher than sham group (P<0.05),and the normal group was higher than model group (P<0.01).Conclusion The depression-like behaviors of WKY depression models were improved by electroacupuncture treatment and the mechanism may be related to the increased expression of GluR1 in hippocampus.
