人皮肤成纤维细胞的分离和原代培养

         

摘要

目的:采用实时细胞监测技术(RTCA)观察人皮肤成纤维细胞增殖过程。方法采集5例患者的皮肤组织,经胰酶消化和传代,对分离纯化后的成纤维细胞使用 RTCA动态检测其增殖过程,并进行免疫荧光检测特异性蛋白表达。结果 RTCA的实验数据比较真实地接近细胞实际的生长情况;免疫荧光结果显示分离的成纤维细胞能够表达波形蛋白。结论 RTCA动态检测能为细胞移植提供有力的保障。%Objective To observe the cell proliferation process by RTCA biosensor technology . Methods The skin fibroblasts from 5 patients were digested by trypsin and passed .The RTCA biosensor technology was applied to detect real‐time cell proliferation process .Immunofluorescence staining was used to detect the expression of vimentin protein .Results The growth curve mapped by RTCA biosensor technology was better close to the actual growth situation .Immunofluorescence staining showed that the isolated fibroblasts expressed vimentin .Conclusion The dynamic detection of cell proliferation process by RTCA can provide a strong guarantee for cell transplantation .
