PGC-1α基因在两种类型鸡骨骼肌间的表达差异

         

摘要

The present study was conducted to detect the expression of peroxisome proliferators activated receptor gamma coactivator alpha (PGC-1α) gene in skeletal muscles of chicken, and to analyze the expressional difference between fast-growing and slow-growing chicken breeds by SYBR Green I semi-quantitative RT-PCR. The chicken breeds included one Chinese indigenous chicken breed (Qingyuan partridge chicken, slow-growing) and one introduced chicken breed (Recessive White chicken, fast-growing). Total RNA was extracted from gastrocnemius and breast muscles of chickens of different weeks of ages (0, 2, 4, 6, 8, 10, 12). Using β-actin gene as an inner control gene, the relative PGC-1α mRNA expression levels in skeletal muscles of fast-growing and slow-growing chicken breeds were uniformly treated and calculated according to the fluorescence threshold ( Ct value ). PGC-1α gene was expressed in all development stages in skeletal muscles, and the expression level was the highest at 0 week of age which was significantly higher than that at other developmental stages. The mRNA expression levels of PGC-1α gene in the skeletal muscle of slow-growing breed Qingyuan partridge chicken were higher than those in fast-growing chicken breed Recessive White chicken of same age. PGC-1α mRNA expression in the skeletal mus- cle of the two chicken breeds followed specific developmental patterns, and might be related to the chicken meat quality.%为了检测过氧化物体增殖物激活受体γ辅激活因子1-α(PGC-1α)基因mRNA在鸡骨骼肌组织中的分布以及在速生型隐性白羽鸡和慢生型清远麻鸡品种间的表达差异,采集不同周龄(0、2、4、6、8、10、12)隐性白羽鸡和清远麻鸡的胸肌和腓肠肌组织样,提取总RNA,以β-actin基因为内参基因,进行实时荧光定量PCR.对PGC-1 α基因mRNA进行均一化处理,利用荧光阈值(Ct值)计算PGC-1α基因mRNA在不同骨骼肌组织以及速生型和慢生型鸡品种间的表达量.发现PGC-1α基因在鸡骨骼肌组织不同发育阶段广泛表达,在初生时表达量最高,并显著高于其他各周龄的表达量;相同周龄时,慢生型清远麻鸡骨骼肌中PGC-1α表达量大多高于速生型隐性白羽鸡.表明不同类型鸡骨骼肌PGC-1α mRNA表达具有特定的发育模式,并可能与鸡肉品质存在关联.
