超声波法提取西洋参茎叶中人参皂苷Re工艺的优化

         

摘要

Using methanol as extraction solvent,ginsenoside Re was extracted from the stems and leaves of American ginseng (Panax quinquefolium L.).And the content of ginsenoside Re was detected by high performance liquid chromatography (HPLC).Single factor tests and orthogonal design were applied to study the effects of ultrasonic frequency,ultrasonic time,extraction temperature and material to liquid ratio on the yield of ginsenoside Re so as to optimize the extraction conditions.Results showed that the rank of factors affecting the yield of ginsenoside Re from high to low was ultrasonic temperature,material to liquid ratio,ultrasonic time,and ultrasonic frequency.The optimized extraction conditions were ultrasonic frequency,45 kHz; ultrasonic time,60min; ultrasonic temperature,45 ℃; and material to liquid ratio,mgenseng∶Vmethandanol=1∶20 (g/mL).%以甲醇为提取溶剂,采用超声波法从西洋参(Panax quinquefolium L.)茎叶中提取人参皂苷Re,通过高效液相色谱法测定人参皂苷Re含量,设计单因素试验和正交试验考察超声波频率、超声波时间、提取温度和料液比对人参皂苷Re含量的影响,优化提取工艺条件.结果表明,各因素对西洋参茎叶中人参皂苷Re提取率的影响由大到小依次为提取温度、料液比、超声波时间、超声波频率.优化的提取条件为超声波时间60 min、提取温度45℃、超声波频率45 kHz、料液比m西洋参∶V甲醇=1∶20(g/mL).
