大豆苷元对慢性应激抑郁大鼠行为学及海马部脑源性神经生长因子表达的影响

     

摘要

目的 探讨大豆苷元抗抑郁作用及其机制.方法 建立大鼠慢性不可预测应激模型,通过强迫游泳(foced swimming test,FST)、敞箱实验(open field test,OFT)观察应激后各组大鼠行为学的变化.运用Western bloting观察经药物治疗后各组大鼠海马齿状回部脑源性神经生长因子(brain-derived neurotrophic factor,BDNF)的改变.结果 经慢性应激14 d后,模型组FST、OFT行为学评分下降,与正常组比较差异有统计学意义(P<0.05),1,3,10 mg·kg-1大豆苷元和丙米嗪治疗14 d后,大鼠FST、OFT行为学评分明显改善(P<0.05).停药7 d后,丙米嗪组、大豆苷元各剂量组FST、OFT评分较模型组均差异无统计学意义(均P>0.05).同时,在海马齿状回部发现,慢性应激可使BDNF蛋白表达下降,经丙米嗪和大豆苷元治疗后BDNF含量均增加.停药7 d后,丙米嗪组、大豆苷元各剂量组再次出现BDNF蛋白表达下降.结论 大豆苷元可能通过增加海马齿状回BDNF的表达改善抑郁模型大鼠行为,产生抗抑郁作用.%Objective To explore the antidepressant effects of daidzein and its possible mechanism. Methods The chronic unpredictable mild stress ( CMS ) model was set up in rats and the hehavior changes were investigated through forced swimming test ( FST ) and open field test ( OFT ) after the treatment of daidzein. The western blotting was used to study the changes of BDNF ( brain-derived neurotrophic factor ) by daidzein. Results The behavioral scores of FST and OFT in the CMS model group in 14 d markedly decreased , in comparison to the normal control group ( P<0. 05 ). However,imipramine and daidzein at 1,3, 10 mg · kg-1 improved the behavioral scores of FST and OFT after 14 d ( P<0. 05 ); 7 days after drugs withdrawn, the FST and OFT scores in imipramine and daidzein groups showed no significant difference compared with the CMS model ( P>0. 05 ). Meanwhile ,the expression of BDNF in DG area of hippocampus was reduced by CMS ( P<0. 05 ), which was increased after the treatment of imipramine and daidzein, respectively. 7 days after treatment, the expression of BDNF in imipramine and daidzein group were declined once again compared with the CMS model control( P>0. 05 ). Conclusion Daidzein may exert the antidepressant effects by improving the behavior performance and increasing the BDNF expression in hippocampus of rat model of depression .
