HPLC法同时测定缬沙坦氢氯噻嗪胶囊中缬沙坦和氢氯噻嗪的含量

摘要

Objective: Simultaneous determinafion of Valsartan and Hydrochlorothiazide in Valsartan and Hydrochlorothiazide capsules was conducted by HPLC. Method:ChromatrexC18(200mm×4.6mm,5μm)methanol:water (70:30), flow rate:1ml/min;detec-tion wavelength:230nm; column temperature:25℃. Results: valsartan8-160μg · ml-1, hydrochlorothiazide in the concentration range1.25-25μg · ml-1, with good linear relationship (r=0.9996, r=0.9994);valsartan average recovery was 99.68%, RSD=0.32%, hydrochlorothiazide average recovery was 99.90%, RSD=0.80%(n=6). Conclusion:HPLC is simple and convenient method of op-erating, testing quickly and accurately, and has good repeatability means of detection, rapid determination of valsartan and hydrochlo-rothiazide content in Valsartan Hydrochlorothiazide capsules, is conducive to quality control of Valsartan Hydrochlorothiazide Cap-sules.%目的：运用高效液相色谱法（HPLC）同时对缬沙坦氢氯噻嗪胶囊中缬沙坦与氢氯噻嗪含量进行测定。方法：ChromatrexC18(200mm×4.6mm，5μm)甲醇：水（70：30），流速：1ml/min；检测波长：230nm；柱温：25℃。结果：缬沙坦在8-160μg· ml-1，氢氯噻嗪在1.25-25μg·ml-1浓度范围内，具有较好线性关系（r=0.9996、r=0.9994）；缬沙坦平均回收率为99.68%，RSD=0.32%，氢氯噻嗪平均回收率为99.90%，RSD=0.80%（n=6）。结论：HPLC是一种操作简单方便，检测快速准确，且具有较好重复性的检测手段，可快速测定缬沙坦氢氯噻嗪胶囊中缬沙坦与氢氯噻嗪含量，有利于缬沙坦氢氯噻嗪胶囊的质量控制。
