GSH对MRL/lpr小鼠肾小管上皮细胞TGF-β1表达影响研究

         

摘要

Objective:To investigate the effects of glutathione (GSH) on transforming growth factor beta1 (TGF-β1) mRNA ex-pression in renal tubular epithelial cells of MRL/lpr mice. Methods:Renal tubular epithelial cells of G57BL/6 female mice and MRL/lpr female mice were cultured and passaged to the third generation, respectively. Expression of TGF-β1 mRNA in cells were evaluat-ed after 5 days of culture with or without GSH. Results:TGF-β1 mRNA expression in renal tubular epithelial cells of MRL/lpr fe-male mice was significantly higher than that of G57BL/6 female mice (P<0.05). Expression of TGF-β1 mRNA in renal tubular epi-thelial cells of MRL/lpr female mice cultured with GSH was significantly lower than that without GSH (P<0.05). Conclusions:GSH may result in decreasing TGF-β1 expression in renal tubular epithelial cells of MRL/lpr female mice.%目的:探讨还原性谷胱甘肽(GSH)对MRL/lpr小鼠肾小管上皮细胞内转化生长因子β1(TGF-β1)mRNA表达的影响,了解还原性谷胱甘肽对肾小管上皮细胞中转化生子因子β1表达的影响。方法:传代培养C57BL/6雌性小鼠及狼疮小鼠模型MRL/1pr雌性小鼠肾小管上皮细胞,给予第3代细胞还原型谷胱甘肽药物干预,培养5天后检测细胞内TGF-β1的表达。结果:狼疮小鼠模型MRL/1pr雌性小鼠肾小管上皮细胞内TGF-β1表达明显高于C57BL/6小鼠(P<0.05)。经还原性谷胱甘肽处理的MRL/1pr雌性小鼠肾小管上皮细胞表达TGF-β1明显低于未处理组(P<0.05)。结论:还原型谷胱甘肽干预可有效减少MRL/1pr雌性小鼠肾小管上皮细胞表达TGF-β1。
