上调miRNA-101表达对人肝癌HepG2细胞系增殖的影响

         

摘要

Objective To investigate the effects of inhibiting enhancer of zeste homolog 2( EZH2 ) on cell proliferation by up-regulating the expression of miRNA-101 in HepC2 cells. Methods Synthetic miRNA-101 mimics was transfected into HepG2 cell lines. The growth of transfected HepG2 cells was detected by methyl thiazolyl tetrazolium ( MTT ). The cell cycle was tested by flow cytometry( FCM ). And the expression of EZH2 mRNA was detected by real-time qRT-PCR. Results The cell proliferative activity was significantly lower in the miRNA-101 mimics group than that in the negative control group and blank control group, respectively( all P < 0. 05 ). In the miRNA-101 mimics group, cells increased significantly at G0/G1 phase[ ( 74.71 ±4.6 )% ] while decreased at S phase[ ( 14.57 ± 1.1 )% ,P <0.05 ];And the expression of EZH2 mRNA was down-regulated( P <0.05 ). Conclusion Up-regulated miRNA-101 can inhibit the expression of EZH2, which plays a negative regulation role on cell proliferative activity of hepatic carcinoma.%目的 通过上调人肝癌HepG2细胞株中miRNA-101表达,探讨miRNA-101抑制靶基因EZH2对肝癌细胞增殖的影响.方法 人工合成miRNA-101模拟体并转染至HepG2肝癌细胞株中,利用MTT法检测细胞增殖抑制、流式细胞仪检测细胞周期、实时荧光定量PCR检测EZH2 mRNA表达.结果 miRNA-101模拟体转染组HepG2细胞增殖活性明显低于阴性对照组及空白对照组(P<0.05),miRNA-101模拟体组G0/G1期细胞明显增多,为(74.71±4.6)%,S期细胞减少,为(14.57±1.1)%(P均<0.05);miRNA-101模拟体组EZH2 mRNA较阴性对照组和空白对照组表达下降(P<0.05).结论 上调miRNA-101可抑制EZH2基因表达,对人肝癌细胞增殖活性具有负调控作用.
