御寒暖胃膏对慢性萎缩性胃炎修复机制的影响

摘要

目的：研究御寒暖胃膏贴敷胃经穴对慢性萎缩性胃炎大鼠胃黏膜TNF－α、PCNA表达的影响，探讨御寒暖胃膏贴敷胃经穴对慢性萎缩性胃炎癌前病变大鼠胃黏膜损伤修复的作用机制。方法大鼠随机分为正常组、模型组、御寒暖胃膏贴敷胃经穴组、药物对照组，采用综合干预方法复制慢性萎缩性胃炎癌前病变大鼠模型，光镜下观察胃黏膜组织的病理变化，采用酶联免疫分析法测定胃黏膜细胞中肿瘤坏死因子α（ TNF－α）、增殖细胞核抗原（ PCNA）的表达水平。结果与正常组比较，模型组大鼠胃黏膜组织病理学检查提示存在腺体萎缩和一定程度的细胞异型增生，大鼠胃黏膜细胞中TNF－α、PCNA表达水平明显升高（ P＜0．05）；与模型组比较，御寒暖胃膏贴敷胃经穴组和药物对照组大鼠胃黏膜组织的病理损伤得到明显修复，大鼠胃黏膜细胞中TNF－α、PCNA的表达水平明显降低（P＜0．05）。结论御寒暖胃膏贴敷胃经穴可以促进CAG大鼠胃黏膜损伤的修复，该作用可能是通过调节胃黏膜细胞TNF－α、PCNA的表达水平来实现的。%Objective To study the effect of Yuhan Nuanwei paste on TNF-αand PCNA in gastric mucosa of chronic atrophic gastritis ( CAG) rat, and to explore the repair mechanisms of Yuhan Nuanwei paste on the stomach meridian on chronic atrophic gastritis and precancerous lesions of CAG rat gastric mucosal injury .Methods The rats were randomly divided into normal group , model group , Yuhan Nuanwei paste on the stomach meridian group and drug control group .By using the comprehensive intervention method to copy precancerous lesions of chronic atrophic gastritis rats'model, the index of the gastric mucosa injury in rats was observed under the naked eye .Then we observed the pathological changes of gastric mucosa tissue using enzyme -linked immunoassay determination of the expression of levels of tumor necrosis factor alpha in gastric mucosa cells (TNF-α), proliferating cell nucleus antigen (PCNA).Results Comparing with the normal group, the model group's rat's gastric mucosa histopathological examination suggested the existence of gland atrophy and a certain degree of cell hyperplasia.The TNF-αand PCNA of rat gastric mucosa cells increased significantly (P<0.05).Comparing with the model group, the pathological damage obviously repaired of the Yuhan Nuanwei paste on the stomach meridian group and the drug control group .The TNF-αand PCNA of rat gastric mucosa cells significantly decreased ( P<0.05) .Conclusion The Yuhan Nuanwei paste on the stomach meridian can promote the CAG rats'gastric mucosa damage repair , and the effect may be through adjusting the TNF-αand PCNA by gastric mucosa cells to come true .
