HILIC色谱柱法检测L-抗坏血酸和D-异抗坏血酸及其在食品分析中的应用

         

摘要

对比了C18柱和HILIC色谱柱对L-抗坏血酸、D-异抗坏血酸的分离效果.进一步考察了在HILIC模式下的流动相组成、柱温和流速对分离L-抗坏血酸、D-异抗坏血酸的影响.实验结果表明:检测波长为245 nm,流动相为0.1 %磷酸+2 g/L甲酸铵-乙腈(10/90,V/V),流速为0.8 mL/min,柱温为20 ℃条件下分离效果最佳,分离度为4.60.采用该方法对三种食品中的L-抗坏血酸、D-异抗坏血酸含量进行测定,三种样品中均有L-抗坏血酸和D-异抗坏血酸检出,含量均在20.59~184.37 mg/100 g之间,相对标准偏差在0.65 %~1.41 %之间.%Comparison of C18column and HILIC column for the separation of L-ascorbic acid and D-isoascorbic acid. The separation conditions, including mobile phase, column temperature and flow rate, were optimized. The results shows that the best wave length was 245 nm, the best mobile phase was 0.1 % phosphoric acid+2 g/L ammonium formate solution-acetonitrile (10:90), the best column temperature was 20 ℃ and the best flow rate was 0.8 mL/min. The optimal resolution was 4.60. This method was further used for ascorbic acid enantiomers determination in three kinds of foods. Both L-ascorbic acid and D-isoascorbic acid were detected in three kinds of foods.The contents were between 20.59~184.37 mg/100 g with reative standard deviation of 0.65 %~1.41 %.
