柔红霉素联合枸橼酸他莫昔芬对人乳腺癌细胞MCF-7的协同作用研究

摘要

目的：研究柔红霉素（Daunorubicin，DNR）和枸橼酸他莫昔芬（Tamoxifen Citrate，TMX）对人乳腺癌细胞MCF-7的协同作用。方法：取对数生长期MCF-7细胞，分为柔红霉素给药组（浓度范围0.25~1.5μmol/L）、枸橼酸他莫昔芬给药组（浓度范围2.5~15μmol/L）和两药混合给药组，采用MTT法考察各给药组作用24h后对MCF-7细胞的增长抑制作用，利用中效原理计算合用指数来判定DNR和TMX的协同作用。结果：联合给药组的细胞抑制率均高于单独给药组，并且IC50时联合给药组的柔红霉素和枸橼酸他莫昔芬剂量低于单独给药组剂量。两药合用时（DNR∶TMX=1∶10，摩尔比），当DNR剂量≥0.188mmol/L，TMX≥1.887mmol/L时，合用指数CI<1，二者表现为协同作用。结论：柔红霉素和枸橼酸他莫昔芬联合应用能抑制人乳腺癌细胞MCF-7生长，在一定条件下，两者为协同作用，并且随着药物剂量增加，效应增强，协同作用呈现更强。%Objective:To study the synergistic effect of daunorubicin combined with tamoxifen on breast cancer cells MCF-7 with the Chou-Talalay method. Methods: The effect of daunorubicin or tamoxifen alone and their combination on MCF-7 cell proliferation were measured by MTT assay. The median-effect principle was used to evaluate the synergistic effect.Results: The combination of daunorubicin and tamoxifen had synergistic effect on MCF-7 cell, when DNR≥0.188mmol/L and TMX≥1.887mmol/L. Conclusion:Daunorubicin combined with tamoxifen has inhibitory effect on MCF-7 cell proliferation, and the interactive effect of the two drugs in combination is synergistic under certain conditions.
