苦瓜多糖含量的苯酚硫酸法检测研究

The conditions for determinating momordica charantia polysaccharide by phenol-sulfuric acid method was studied. The results showed the optimum determinating conditions were 1 mL 1%polysaccharide solution and 0.4 mL 6%phenol, 4.5 mL sulfuric acid color 10 min at 60℃, mensurating absorbance by using spectrophotometer at 493 nm. These procedures improved stability and repeatability of phenol-sulfuric acid method and were easy to operate.%为了寻找适于测定苦瓜多糖含量的稳定方法，本文对苯酚硫酸法测定苦瓜多糖的条件进行了研究。实验结果表明，在苯酚硫酸法中，测定条件为1 mL 1%苦瓜多糖溶液与0.4 mL 6%苯酚、4.5 mL浓硫酸60℃显色10 min，493 nm波长处测定吸光度。结果表明，改进后的苯酚硫酸法是测定苦瓜多糖含量的一种回收率高，稳定性好的简便方法。
