NH3应激肉鸡呼吸道两株致病菌的分离和鉴定

         

摘要

To explore the effect of NH3 stress on broiler breeding and chicken quality,20 one-day-old Ross 308 broilers(10 Male, 10 Female)were selected randomly and exposed to 60 mg/m3NH3. The experiment lasted for 42 days. By the end of the study, 5 broilers were selected randomly, unstimulated whole saliva samples were collected from posterior pharyngeal wall. Microbial in the samples were isolated, cultured and identified by using dilution-plate method and molecular biological technique.The results based on 16S rDNA sequence analysis showed that the isolated strains of TSB87001 and TSB87002 were related to Cronobacter sakazakii(GenBank:HQ880345.1)and Vibrio cholerae(GenBank:JN873349.1),respectively,and both of the them the similarities were all reached to 97 %.Exposed to NH3 environment for long time,which would assist the pathogenic microorganism to invade and planting the birds, then which would cause a series of the food security of broiler origin.%为探讨NH3应激对肉鸡养殖业及鸡肉品质的影响,选用1日龄健康罗斯308肉鸡20羽,公雏和母雏各10羽,饲养于浓度为60 mg/m3NH3环境中,试验期为42 d.试验结束,随机挑选5羽试验肉鸡,用无菌棉拭子于咽后壁取样,利用稀释平板法对呼吸道微生物进行分离培养,并结合分子生物学技术对分离得到的微生物进行初步鉴定.结果显示:从NH3应激肉鸡呼吸道中分离得到的菌株TSB87001和TSB87002分别与Cronobacter sakazakii(GenBank:HQ880345.1)和Vibrio cholerae(GenBank:JN873349.1)的相似性达到97 %,初步鉴定为阪崎肠杆菌和霍乱弧菌.长时间的NH3刺激有助于病原微生物的侵入和在动物机体内定植,引发鸡肉产品的一系列食品安全问题.
