鸡血藤不同成分抗AA诱导的血小板聚集作用的实验研究

         

摘要

Objective To screen the effective parts of spatholobus suberectus on anti-platelet aggregation. Methods Overall composition of spatholobus suberectus (OCSS) were extracted by water extraction and alcohol precipitation,then total flavonoids (TF-SS) and nonflavonoids parts (NFPSS) in overall composition of spatholobus suberectus were separated by AB-8 macroreticular resin. Healthy SD rats divided into 11 groups were given a high,medium,low dose of OCSS,TFSS,NFPSS by intragastric administration in-dividually for 2 days,and the maximum aggregation rate of platelet (MARP) induced by arachidonic acid (AA) were detected. Results The MARPs in positive group,overall composition group and total flavonoids group(except the low dose group) were significantly low-er than those in negative group (P<0.01),while there was no significant difference between nonflavonoids part group and negative group(P>0.05). Conclusion Flavonoids is the effective part of spatholobus suberectus on anti-platelet aggregation.%目的:筛选由花生四烯酸(AA)诱导的鸡血藤抗血小板聚集作用的有效成分。方法采用水提醇沉法,得鸡血藤浸膏(总成分),用AB-8大孔吸附树脂提取其中的总黄酮成分,提取黄酮后剩下的滤液即为非黄酮成分。实验分组:分为鸡血藤浸膏组、总黄酮成分组、非黄酮成分组、阿司匹林对照组、生理盐水对照组。分别给SD大鼠灌胃高、中、低不同剂量的鸡血藤各组分。测定由AA诱导的血小板5min之内的最大聚集率。结果鸡血藤浸膏高、中、低剂量各组的聚集率(%)分别为:4.6±1.1、8.3±2.0、42.5±4.2;鸡血藤总黄酮成分高、中、低剂量各组的聚集率(%)分别为:6.4±1.5、37.6±3.5、70.1±4.3;鸡血藤非黄酮成分高、中、低剂量各组的聚集率(%)分别为:65.7±3.2、69.0±3.8、72.5±4.4;阿司匹林对照组的聚集率(%)为:41.6±3.8;正常对照组的聚集率(%)为:70.3±4.6。鸡血藤浸膏组、鸡血藤总黄酮成分组(低剂量组除外)、阿司匹林组分别显著性低于正常对照组(P<0.01)。鸡血藤非黄酮成分高、中、低剂量各组与生理盐水对照组比较,结果无显著性差异(P>0.05)。结论鸡血藤总黄酮具有显著性的抗AA诱导的血小板聚集作用,为开发新的抗血小板聚集药物打下基础。
