通心络超微粉对缺血性脑卒中大鼠微血管新生影响的实验研究

         

摘要

目的 探讨通心络超微粉对缺血性脑卒中大鼠微血管新生及相关因子的影响.方法 利用开颅结扎法阻断大鼠一侧大脑中动脉制作局灶性脑缺血模型(MCAO),筛选后随机分为假手术组、模型组、通心络大、中、小剂量组、尼莫地平组,给药14 d后,免疫组化方法测定MCAO大鼠微血管密度(CD31)、血管内皮生长因子(VEGF)表达,ELISA放射免疫方法和硝酸还原酶法检测血清VEGF、一氧化氮(NO)的水平.结果 通心络大、中剂量组、尼莫地平组CD31蛋白阳性分布密集且着色明显,均强于模型组;通心络大剂量组VEGF表达明显加强,优于模型组.通心络大、中剂量组大鼠血清NO水平明显提高优于模型组(P<0.05,P<0.01),且通心络大剂量组优于通心络中剂量组(P<0.05);通心络大剂量组大鼠血清VEGF水平明显提高优于模型组(P<0.01),通心络大剂量组优于通心络小剂量组和尼莫地平组(P<0.05,P<0.01).结论 通心络能够明显增加缺血侧脑组织微血管密度,促进血管生长因子VEGF、NO表达,具有促进微血管新生的作用.%Aim To explore the interventional effects of Tongxinluo supermicropowder on angiogenesis , related angiogenesis factors and their expression. Methods With craniotomy ligation, a side of the middle cerebral artery of rats were blocked to establish the model of focal cerebral ischemia ( MCAO ). After filtration, male SD rats were randomly divided into sham - operation group, model control group, TXL-H, TXL-M, TXL-L and Nimodipine. Microvascular density of cerebral cortex ischemia area and related angiogenesis fac -tors were tested through immunohistochemical kit at 14 d after heart perfusion fixation. The levels of NO, VEGF in serum were tested through specific kit at 14 d after conventional separation of serum. Results CD31 expression of Tongxinluo high-dose, middle-dose and Nim groups were intensive, while sham group was lightly dyed. In treatment groups, Tongxinluo high-dose group was most strongly dyed. The expression ofVEGF was slightly positive in sham group , but increased obviously in Tongxinluo high -dose group at 14 d after focal ischemia, compared with model group. The levels of NO in serum of Tongxinluo high-dose and middle-dose group were significantly increased , compared with model control group (P < 0.05, P < 0. 01) , and Tongxinluo high-dose group was more significant ( P < 0. 05 ). The contents of VEGF in serum of Tongxinluo high-dose group were also significantly increased, compared with model control group , Tongxinluo low-dose and Nim group ( P < 0. 05, P < 0. 01). Conclusion Tongxinluo can significantly improve the microvascular density of ischemic stroke rats and increase the levels of NO , VEGF expression. Tongxinluo shows obvious protective effects on angiogenesis .
