温度对DEN-2型病毒在白纹伊蚊体内外潜伏期的影响

         

摘要

To investigate the effect of incubation temperature on extrinsic incubation period, Aedes albopictus were oral ly infected with dengue virus type 2 suspension, then fully blood engorged mosquitoes were separated and transferred to new cartons and retained at 18 °C , 21 °C , 26 °C , 31 °C and 36 °C with 10% sucrose. Mosquitoes were sacrificed on 3 d, 4 d, 5 d, 7 d, 10 d, 14, 20 d and 25 d post infection. Immunofluorescence assays were used to detect antigen of dengue virus type 2 in the head, salivary gland and thorax abdomen. Virus in thorax abdomen was detected on 25 d,7 d,5 d,4 d and 3 d post infection. The extrinsic incubation period was 10 d,7 d,4 d and d4 separately, when mosquitoes were maintained at 21 °C ,26°C ,31°C and 36°C. But virus in salivary was not detected during a 25 day incubation period when mosquitoes were retained at 18°C. It was suggested that the extrinsic incubation period of A. Albopictus shortened gradually with the increase of extrinsic incubation tem perature.%目的 研究温度对登革2型病毒在蚊体内的外潜伏期的影响.方法 登革2型病毒人工经口感染白纹伊蚊,冻麻挑出吸饱血的雌蚊放入新的蚊笼,分别置于18℃、21℃、26℃、31℃和36℃中饲养,第3 d、4 d、5 d,7 d、10 d、14 d、20 d、25 d分别收集并解剖白纹伊蚊,以间接免疫荧光法分别检测蚊虫头部、唾液腺和胸腹部的登革2型病毒抗原.结果 置于18℃、21℃、26℃、31℃和36℃饲养的白纹伊蚊分别在第25 d、7 d、5 d、4 d、3 d时胸腹部中开始检出阳性.21℃、26℃、31℃和36℃饲养的蚊虫唾液腺阳性检出的时间分别为10 d、7 d、4 d、4 d,18℃在感染后的25 d内,唾液腺均未检出阳性.结论随着温度的升高,登革2型病毒在白纹伊蚊体内外潜伏期逐渐缩短.
