2010年福建省肠道病毒71型分离株的基因型特征研究

         

摘要

目的 全面研究和了解肠道病毒71型在福建省的遗传背景,分析和探讨该型病毒在我省的地理分布特征及传播特征.方法 对2010年福建省9个设区市的50株EV71分离株进行VP1区全长基因序列测定,并对核苷酸序列进行同源性比较及遗传进化分析.结果 2010年福建省50株EV71型分离株的VP1区核苷酸序列全长均为891 bp,核苷酸及氨基酸序列同源性比较,50株EV71型分离株之间的VP1区核苷酸序列同源性为95.4%~100%,编码蛋白氨基酸序列同源性为97.3%~100%,与2008年阜阳流行株Fuyang17.08-2同源性最高,核苷酸序列同源性为97.1%~99.3%,氨基酸序列同源性为98.7%~100%;VP1区基因遗传进化分析,与Fuyang17.08-2株进化关系较为接近,同属于C4a基因亚型.结论 2010年福建省50株EV71型分离株均属于C4a基因亚型,未产生明显的抗原漂移及变异;福建省及各个地市均分布亲缘关系较近的C4a基因亚型的EV71多传播链病毒,应加强长期动态地监测和分析.%Enterovirus type 71 (EV71) is a common cause of pediatric hand, foot and mouth disease (HFMD). The genetic background of current EV71 isolates was investigated, and the distribution and transmission characteristics of EV71 strains in Fujian Province were analysed in this study. The complete VP1 sequences of 50 EV71 strains, isolating from specimens of 50 HFMD patients in Fujian in 2010, were sequenced, homologically compared and phylogenetically analysed. Sequences analysis revealed that the full-length VP1 region of 50 Fujian EV71 strains was 891bp. Within 50 Fujian EV71 isolates, the nucleotide and amino acid identity were 95. 4%-100% and 97. 3%-100% respectively, sharing high homology with Fuyang 17. 08-2 both in nucleotide and amino acid sequences, with the identity of 97. L%-99. 3% in nucleotides and 98. 7%-100% in amino acids, respectively. Phylogenetic analysis based on complete VP1 sequence also indicated that 50 Fujian EV71 strains had a close genetic relationship with Fuyang 17. 08-2, which also belonged to the same subgenotype, C4a. Therefore, no obvious antigenic drift or mutation was observed. However, as closely related subgenotype C4a EV71 have multiple transmission chains in Fujian Province and in each prefecture, extensive surveillance and analysis are required.
