HSP 70表达与雄激素依赖性和非依赖性前列腺癌的关系

摘要

目的 探讨热休克蛋白70(HSP70)与雄激素非依赖性前列腺癌(PCa)发生的关系。 方法 采用免疫组化方法测定2例正常前列腺组织、20例雄激素依赖性PCa和14例雄激素非依赖性PCa石蜡包埋标本HSP70的表达。 结果 HSP70在正常前列腺基底细胞中呈阳性表达,分泌细胞中为阴性表达。20例雄激素依赖性PCa中16例(80.0%)无HSP70表达,3例有微弱表达,1例为异质性表达。14例雄激素非依赖性PCa中13例(92.8%)呈弥漫、高水平的HSP70表达,1例为阴性,与雄激素依赖PCaHSP70表达率的比较,差别有极显著性意义(P<0.01)。 结论 HSP70过表达与雄激素依赖性PCa向雄激素非依赖性PCa的进展有关。%Objective To investigate the relationship between major humanheat shock protein 70(HSP 70) and the development of androgen-independent prostate cancer(PCa). Methods The formalin-fixed, paraffin-embedded samples of normal prostate tissue(2cases), androgen-dependent PCa(20cases) and androgen-independent PCa(14 cases) tissue were evaluated immunohistochemically for HSP 70 expression. Results In normal prostate tissue, basal cells exhibited positive HSP 70 staining, whereas secretory cells were negative. HSP 70 was undetectable in 16 cases(80%) of androgen-dependent PCa and weak in 3 cases,heterogeneous in 1 case(20%). In contrast,13 cases(92.8%) of androgen-independent PCa displayed diffuse and high level HSP 70 staining whereas negative expression in 1 case. HSP 70 expression level of androgen-independent PCa tissue was higher than that of androgen-dependent PCa with statistically very significant difference(P<0.01). Conclusions The result indicates that HSP 70 overexpression in PCa is correlated with the progression of PCa from androgen dependence to androgen independence.
