彩色马蹄莲胚拯救技术研究

         

摘要

以彩色马蹄莲栽培品种‘M7507’、‘M2’、‘M37’的幼胚为实验材料,采用L9k(34)的正交实验设计,对其离体胚培养,即胚拯救的适宜发育阶段和适宜培养基进行研究.结果表明:(1)幼胚发育阶段显著影响彩色马蹄莲胚拯救成功率,最佳发育阶段幼胚的形态特征为球状、乳白色、光滑; (2)适宜诱导培养基为MS+6-BA 1.0 mg/L+NAA 0~0.2mg/L; (3)幼胚接种后不同品种初始萌发所需时间不同,‘M2’、‘M7507’、‘M37’依次为14、16、21d,品种间萌发率经新复极差法检验差异不显著;(4)不同品种的果实膨大速度不同,发育到胚拯救的最适阶段,‘M7507’、‘M2’、‘M37’分别需要50、60、65 d左右的时间.%The young embryo of coloured Zantedeschia hybrids' M7507', 'M2', 'M37'as the experimental materials, the orthogonal experimental design of L9(34)was adopted to study the optimal media and development stage for embryo rescue. The results showed that:(1)The development stage of young embryo was the key factor affecting embryo rescue, and the young embryo was ball, smooth, and milk white after 50 d or bo pollinated, was the best development phase;(2)MS+6-BA 1.0mg/L+NAA 0~0.2 mg/L was the best induction media.(3)The time of different varieties to germination was different, and that for'M2','M7507', 'M37'was 14 d, 16 d, 21 d, respectively. (4)The growth rate of fruits of different varieties was different, and that for 'M7507', 'M2', 'M37' was 50 d, 60 d, 65 d, respectively to the best development phase for embryo rescue.
