交泰丸对糖尿病大鼠尿液代谢谱的影响

         

摘要

目的:通过尿液代谢组学的检测,探讨交泰丸对糖尿病大鼠尿液代谢谱的影响.方法:SD大鼠高脂高糖饲料喂养6周后,腹腔注射链脲菌素(Streptozotocin,STZ) 37 mg/kg,建立2型糖尿病模型,正常对照组注射相同剂量的柠檬酸缓冲液.检测1周后模型组大鼠空腹血糖值FBG≥7.0 mmol/L为糖尿病模型大鼠.糖尿病大鼠随机分为正常对照组6只、模型组6只、交泰丸组6只.用药6周后收集尿液.采用氯甲酸乙酯(ECF)衍生和气相色谱质谱联用(GC/MS)的方法检测大鼠尿液代谢谱的变化.结果:交泰丸治疗6周后,模型组和交泰丸组的大鼠代谢模式图分开,同时正常组与交泰丸组的大鼠代谢模式图较接近,其结果与常规生化形态基本一致.结论:交泰丸能够改善糖尿病大鼠尿液代谢谱,对糖尿病大鼠具有治疗作用.%Objective:To investigate the effect of Jiaotai Pill on urinary metabolic spectrum in diabetic rats by detecting urine metabolomics.Methods:SD rats feed with food containing high fat and high sugar for 6 weeks,and then established the model of diabetes Ⅱ by intraperitoneal injection of Streptozotocin (STZ,37 mg/kg),control group was intraperitoneal injection with the same dose of citrate buffers.After one week,the rats with fasting blood glucose level FBG ≥ 7.0 mmol/L were chosen as diabetes models.The rats was divided into three groups:control,model,Jiaotai Pill,each group contained 6 rats,rats in Jiaotai Pill group treated with Jiaotai Pill for 6 weeks,then collected the urine of rats in each group.The changes of urinary metabolic spectrum in rats of each group were measured by ethyl chloroformate (ECF) and gas chromatography-mass spectrometry (GC/MS).Results:Jiaotao Pill treated for 6 weeks,the metabolic pattern was difference between model group and Jiaotai Pill group,and at the same time,the metabolic pattern was no difference between normal control group and Jiaotai Pill group,this result was same to normal biochemical patterns.Conclusion:Jiaotai Pill can improve the urinary metabolic spectrum of rats with diabetes,and has therapeutic effect on diabetes.
