鼻咽清颗粒对鼻咽癌CNE-2细胞周期和凋亡的影响及可能机制

         

摘要

目的 探讨鼻咽清颗粒对鼻咽癌CNE-2细胞周期、凋亡影响及其可能作用机制.方法 CNE-2细胞Hoechst 33342-PI双染后,在荧光倒置显微镜下观察细胞形态,流式细胞仪检测(flow cytometry FCM)检测细胞周期和凋亡考察鼻咽清颗粒对CNE-2细胞周期和细胞凋亡的影响,West-blot考察鼻咽清颗粒对CNE-2细胞Caspase-3和Bcl-2蛋白表达的影响.结果 与阴性对照组相比,随着鼻咽清颗粒浓缩液浓度增加,细胞凋亡率增加,G1的比例增加,West-blot实验结果表明,随着培养基中鼻咽清主药浓度的增加,促进细胞凋亡的Caspase-3蛋白表达增加,抑制细胞凋亡Bcl-2蛋白表达降低.结论 鼻咽清颗粒能诱导caspase-3表达,下调Bcl-2蛋白表达,将CNE-2细胞阻滞于G1期,诱导细胞凋亡.%Objective To investigate the influence and p-otential machnism of Bi-Yan-Qing Granula on cell cycle distribution and apoptosis of nasopharyngeal cancinoma cell CNE-2. Methods CNE-2 cell apoptosis was observed by Hoechst-PI double staining;while cell cycle distribution and apoptosis was evaluated by flow cytometry ( FCM ),West-blot was uesed to determine the experess of Caspase-3 and Bcl-2. Results With the increase of the concentration of Bi-Yan-Qing solution, apopotosis and G1 phase increase compared with control group (P<0.05),West Blot showed after treatment with Bi-Yan-Qing Granula concentrated solution caspase-3 protein expression increased,while Bcl-2 protein expression decresed. Conclusions Bi-Yan-Qing Granula can promote apoptosis and G1 Cell cycle arrest of nasopharyngeal carcinoma cell line CNE-2 by inducing expression of caspase-3 protein as well as inhibiting Bcl-2 protein expression.
