Protein splicing is post-translational process mediated by inteins. Inteins are internal protein sequences that self-excise and splice together the flanking sequences with a peptide bond. The discovery of inteins enhances theories of gene expression and post-translation, protein splicing technology can be applied in protein research and protein engineering. This review provides a survey of recent applications of protein splicing including protein labeling, protein segmental isotope-labeling, protein cyclization, protein chip, gene therapy and so on.%蛋白质剪接(protein splicing)是由蛋白质内含子介导的在蛋白质水平上翻译后的加工过程.蛋白质内含子(intein)是指前体蛋白中的一段插入序列,它在蛋白质翻译后的成熟过程中能自我催化,使自身从前体蛋白中切除,并将其两侧的多肽片段以肽键连接,形成成熟的蛋白质.蛋白质内含子的发现丰富了基因表达和蛋白质翻译成熟过程的理论,而且在蛋白质研究和蛋白质工程中有广泛的应用.本文试图综述蛋白质剪接在蛋白质特异位点标记、蛋白质片段化标记同位素、蛋白质环化、蛋白质芯片、基因治疗等研究中的应用.
