Objective: To investigate the effect of autoantibodies against α1-adrenergic receptor on the expression of Transforming growth factor-β1 (TGF-β1) in diabetic wistar rats and its mechanism of tubulointerstitial lesions were studied. Method: 64 Wistar rats were divided into blank control rat group and the diabetic rat group. Blank control rat group and diabetic rat group were divided into non-mediated group and the autoantibodies against α1-adrenergic receptor-mediated group. The experimental diabetic rats were yielded by injecting streptozotocin. Blood glucose was measured by glucose oxidase. Serum creatinine was measured by radioim-munoassay. The expression of TGF-β1 in tubular ground substance was exanmined by immunohistochemistry. Results: Diabetic rats were model-inged successively. The TGF-β1 was exanmined in renal cortex distal tubule in both blank control rat group and diabetic rat group mediated by autoantibodies against α1-adrenergic receptor and the TGF-β1 expression level in diabetic rat group was significantly higher than that in blank control rat group(P<0. 01). Conclusion: The expression of TGF-β1 in diabetic rats which mediated by autoantibodies against α1 -adrenergie receptor is increased than that in healthy rats. The expression of TGF-β1 is related to autoimmune.%糖尿病(Diabetes Mellitus,DM)最常见的并发症之一是糖尿病肾病( Diabetic Nephropathy,DN),主要表现为肾间质纤维化,以肾小管萎缩和退变、细胞外基质( Extracelluarmatrix,ECM)聚集为主要病理改变[1].α1-肾上腺素受体抗体(简称α1-受体抗体)对应的受体属于G蛋白偶联受体(G-Protein-Coupled-Receptor,GPCR)家族,可参与肾脏功能及免疫调节应答[2].Gouwy等[3]研究发现,自身受体受到反复刺激后可通过内化机制产生自身受体抗体而引起病理效应.转化生长因子-β1 (Transforming Growth Factor-β1,TGF-β1)是一组由多种细胞产生的多肽生长因子,可导致ECM蛋白表达增加并抑制其降解[4,5],但其在α1-受体抗体介导下引起的ECM重构尚未见报道.为了探讨α1-受体抗体是否可通过调节TGF-β1,最终导致肾基质重构,本研究建立DM大鼠模型,观察α1-受体抗体介导的TGF-β1在DM大鼠肾小管的表达及其促进ECM积聚和肾基质重构的作用.
