温脾实肠方对脾阳不足型泄泻动物外周血T淋巴细胞亚群及肠粘膜微观结构的干预

摘要

Objective:To Observe the effection of wenpishichang formula on T-lymphocytes subgroup(CD4,CD8,CD4/CD8)and Structure of intestinal mucosa in mice with splcen-yang deficiency diarrhea. Methods: Healthy adult ICR mice . male and female in half. were randomly divided into four groups(normal control group,model control group,fuzilizhong Pill group,high dose group,middle dose group and low dose group), 10 rats in each group. The spleen-yang deficiency diarrhea model of mice were induced by dahuang decoction.One hour after the last treatment,eye blood samples were collected in each groups.The T-lymphocytes subgroup were detected by FCM and the Structure of intestinal mucosa were detected by Light microscope.Results: The levels of CD4 and CD4/CD8 in mice with spleen-yang deficiency diarrhea were lower than the normol control group.The large doses of wenpishichang formula can raise the level of CD4 and CD4/CD8 .The Structure of intestinal mucosa did not have significant changes. Conclusion: Wenpishichang formula can correct the level of T-lymphocytes subgroup in mice with spleen-yang deficiency diarrhea.%目的:观察温脾实肠法组方对脾阳不足泄泻模型小鼠外周血中T淋巴细胞亚群(CD4、CD8、CD4/CD8)的影响及肠粘膜微观结构的干预,探讨温脾实肠颗粒治疗脾阳不足泄泻的机理.方法:方法:取ICR小鼠,随机分为正常对照组,模型对照组、温脾实肠颗粒大中小剂量组和附子理中丸组.采用苦寒泻下法复制脾阳不足动物模型.造模各组均灌胃给予100%大黄水提液(0.3ml/10g,1次/d)同时给予药物连续14日.末次给药1h,眼眶静脉丛取血,流式细胞仪检测CD4、CD8、及CD4/CD8.另取小鼠回肠组织,光镜观察温脾实肠法组方对脾阳不足型泄泻小鼠肠粘膜结构的影响.结果:脾阳不足泄泻模型组小鼠外周血中CD4和CD4/CD8均较正常组下降(p<0.05).给予药物治疗后,温脾实肠大剂量组和中剂量组可明显升高外周血中CD4,温脾实肠人剂量组可明显升高外周血中CD4/CD8比值,与模型对照组比较具有显著性差异(p<0.05,P<0.01).光镜观察温脾实肠法组方对脾阳不足型泄泻小鼠肠粘膜上皮细胞束见明显病理学改变.结论:温脾实肠方通过对脾阳不足泄泻小鼠外周血中T细胞亚群的调节而改善脾虚泄泻.
