目的 研究脯氨酸丰富的肌醇多磷酸5-磷酸酶(PIPP)在组氨酸557位点突变为丙氨酸时PIPPH557A对小鼠受精卵卵裂的调节作用.方法 将HA-PIPPH557A重组质粒转染到小鼠受精卵中,用免疫荧光染色法检测HA-PIPPH557A对小鼠受精卵卵裂的影响.然后使用相差显微镜观察并计数HA-PIPPH557A对受精卵卵裂率的影响.结果 HA-PIPPH557A在小鼠受精卵的有丝分裂过程中对卵裂的调节与PIPP的作用相反.并且他们之间的比较具有显著意义(P<0.001).结论 PIPPH557A在小鼠受精卵中通过调节PIPP的活性作用干扰受精卵卵裂.%Objective To study tie regulative role of PIPPH557A (Proline-rich Ihositol Polyphosphate 5-Phosphatase H istidine 557 A lanine) on cleavage of mouse fertilized eggs.M ethods With respective transfection of HA-PIPPH557A ,HA-PIPP or HA vector to mouse fertilized eggs to detect the division rate by direct counting under dissectm icroscope and indirect in m unofluorescence anairsis under fluorescence microscope. Results Contrary effects of HA-PIPP with HA-PIPP on cell cleavage w ere observed duringmitot-ic course of mouse feitilized eggs .The division rate was significant, com pared with each other (ρ<0 .05) .Conclusion PIPPH557A may interruptted cell clavage of mouse fertilized eggs by regulating the activity of PIPP .
