鸡胚发育过程GFP标记视顶盖神经元迁移规律及NF分布特点的研究

         

摘要

目的:阐明鸡胚发育过程视顶盖层的组织学结构,神经元迁移的模式及神经丝蛋白的分布特点.方法:鸡胚正常培养到第3天(E3),采用鸡胚带壳开窗培养方法,取出3~4 ml蛋清,开窗培养至E5,将质粒pCAGGS-GFP通过活体原位电转基因技术导入视顶盖,电转后继续培养到胚胎发育的E8~E12,取材、固定、包埋、切片后进行荧光免疫组化分析不同时期各层结构的形成和神经丝蛋白的分布.结果:在鸡胚发育过程,视顶盖从内向外最终共形成6层结构,大部分神经元垂直于视顶盖平面迁移至视神经纤维层终止,部分细胞沿视神经纤维层继续水平迁移,在中央灰质层(SGC)层,细胞迁移复杂,既存在纵向也存在横向迁移模式,NF的表达具有明显层的特点.结论:鸡胚发育过程视顶盖神经元不但存在从内向外垂直迁移的模式,而且还存在水平横向迁移的模式.%Objective: To clarify the optic tectum layer histological structure, neuron migration mode and neurofilament protein espression characteristic during chicken embryonic development. Methods: Fresh eggs were cultured in normal condition for three days ( E3) , and 3 -4 ml egg white was removed from egg, then continuing culture to E5. pCAGGS-GFP plasmid was injected into the optic tectum and then handled by in vivo electroporation technology. Tissues were obtained from E8 to E12 and fixed with 4% PFA, embeded with OCT, and cut into frozen sections. Fluorescence immunohistochemistry method was used to analyze the different periods of the layers forming and neurofilament protein distribution. Results:The chicken embryo optic tectum structure could be divided into six layers from inside to out, most of the neurons vertically migrate to the termination of the stratum opticum layer, some of the cells move along as the fiber layer lateral migration. In the SGC layer cell migration complex, both of the longitudinal and transverse migration modes exist. The expression of the neurofilament protein having a significant layer characteristics. Conclusion: Tectum neurons have not only longitudinal migration mode from the inner, but also existence of horizontal migration mode.
