Objective To explore the possible mechanism of leptin on human SK-BR-3 breast cancer cells, cell proliferation and vascular endothelial growth factor ( VEGF) expression. Methods SK-BR-3 cells were treated with different concentration of leptin for different hours in vitro, respectively. Cell proliferation was evaluated by CCK-8 method. The expression of VEGF mRNA and protein were detected in SK-BR-3 by real time PCR and western blot, respectively. Results Cell proliferation of SK-BR-3 was in a dose and time-dependent way in vitro (P < 0. 05 ). The expression level of VEGF mRNA and protein were increased significantly in a leptin dose-dependent manner ( P < 0. 05 ). Conclusions Leptin promotes the growth of human SK-BR-3 breast cancer cells in vitro, which plays a role in the mechanism of angiogenesis and metastasis by increasing the expression of VEGF.%目的 观察瘦素对人乳腺癌SK-BR-3细胞增殖及对VEGF表达的影响,探讨其可能机制.方法 不同浓度瘦素与人乳腺癌SK-BR-3细胞共同培养后,CCK-8法检测其对人乳腺癌SK-BR-3细胞增殖的影响,实时荧光定量PCR法及Western印迹法检测VEGF mRNA及蛋白表达情况.结果 在一定范围内,瘦素呈时间、剂量依赖性促进SK-BR-3细胞的生长(P<0.05);细胞受不同浓度瘦素作用后VEGF mRNA及蛋白表达量明显增加并具有剂量依赖性(P<0.05).结论 在体外瘦素对人乳腺癌SK-BR-3细胞有明显增殖作用,通过促进VEGF的表达,在促进血管的生成及肿瘤转移机制中可能发挥一定作用.
