抗戾解毒汤体内抑制小鼠高致病性禽流感病毒 H5N1增殖

         

摘要

Objective To analyze and evaluate the in vivo effect of the anti influenza herbal decoction to reduce proliferation of highly pathogenic H5N1 avian influenza virus.Methods After anesthetized,the mice were infected intranasally with mouse adapted highly pathogenic H5N1 avian influenza virus.And within 120 minutes after infection,the experimental group were treated with intragastric adminis-tration of anti influenza herbal decoction;Oseltamivir group injected Oseltamivir liquor intraperitoneally;control group treated with PBS buffer intragastric administration .Then weight change and survival time were monitored .Real time PCR was used to measure viral load in the lungs.Results The survival rate of the experimental group was 80%,was obviously higher than that of Oseltamivir group (53.3%) or con-trol group(3.3%)(P<0.01).The decreased weight of experimental group was also lower than that of other two groups .There was not sig-nificant difference in the viral load level in the lung between experimental and Oseltamivir group (P>0.05),that of experimental group was significantly lower than that of control group (P<0.01).Conclusions The anti influenza herbal decoction has a strong effect on reducing proliferation of highly pathogenic H5N1 avian influenza virus,may potentially serve as a novel,highly effective anti influenza drug.%目的:探讨抗戾解毒汤体内抑制高致病性禽流感病毒H5 N1增殖的效果。方法小鼠麻醉后使用高致病性禽流感病毒 H5 N1的鼠适应株滴鼻以感染,感染后120 min内,实验组给予抗戾解毒汤灌胃,奥司他韦组给予奥司他韦溶液腹腔注射,对照组给予PBS缓冲液灌胃,之后观察体重变化和死亡率,并通过实时PCR方法检测肺部病毒负载水平。结果实验组存活率(80%)较奥司他韦组(53.3%)和对照组(3.3%)显著提高(P<0.01),实验组体重下降也低于其他两组;实验组肺部病毒负载水平较对照组显著降低(P<0.01),和奥司他韦组无明显差异(P>0.05)。结论抗戾解毒汤对感染H5 N1病毒的小鼠的病毒增殖抑制作用明显,且保护作用强,可以作为流感病毒的备选或者辅助药物。
