Objective To explore the level of programmed death-1 (PD-1) on the CD8 + T cell surfaces in the peripheral blood between the gastric cancer patients and the healthy controls, and the relationship of the level of PD-1 with the cancer progress. Methods There were 66 cases of gastric cancer patients in various cancer types, and 34 healthy controls to obtain the blood samples and isolate PBMCs. The positive expression of the PD-1 on the CD8 + T cell surfaces in the peripheral blood was detected by the flow cytometry. Results The positive expression of PD-1 on the CD8 + T cell surfaces in the peripheral blood of the gastric cancer patients was higher than that of the healthy controls (17.81% ± 4.74% vs 8.22% ±5.17% , P < 0. 0001 ). The expression of the PD-1 on the CD8 + T cell surfaces in the peripheral blood of the various cancer patients were different, the difference had statistical significance. Conclusion The positive expression of PD-1 on the CD8 + T cell surfaces in the peripheral blood of the gastric cancer patients is higher than that of the healthy controls, and it is related with the various gastric cancer types.%目的 观察胃癌患者及健康体检者外周血CD8+T细胞表面程序性死亡受体-1 (programmed death1,PD-1)的表达水平,及其与胃癌病变程度的关系,探讨其临床意义.方法 不同分期的胃癌患者66例,健康体检者34例;取新鲜血液分离外周血单核细胞(peripheral blood mononuclear cells,PBMCs),运用流式细胞仪(flow cytometry,FCM)检测外周血CD8+T细胞表面PD-1表达的阳性.结果 对66例胃癌患者及34例健康体检者外周血进行流式细胞仪分析,结果显示胃癌患者外周血CD8+T细胞表面PD-1阳性表达明显高于健康体检者,差异有统计学意义(17.81%±4.74% vs 8.22%±5.17%,P<0.0001),且在不同分期胃癌患者外周血CD8+T细胞表面PD-1表达的不同,差异均有统计学意义.结论 胃癌患者外周血CD8+T细胞表面PD-1的表达较健康体检者明显上调,且与胃癌的不同分型呈正相关.
