硝苯地平对葡萄糖、胰岛素刺激的肌细胞内钙流动的抑制作用

         

摘要

Objective To investigate the effect of nifedipine, a commonest calcium antagonist in clinic, on glucose metabolism. Methods Intramyocytic calcium fluxion of rat soleus muscle were measured. Cultured myocytes of rat soleus were loaded with Fluo-3 fluorescence probe, and the changes of intramyocytic Ca2+ concentration were recorded by laser scanning conforcal microscopy. Results 16. 7mmol/L of glucose and/or 100μU/ml of insulin significantly increased the intramyocytic Ca2+concentration, and nifedipine blocked the stimulating effects of glucose and insulin in a doesdependent fashion. In basic situation, nefedine in a low dose (0. 5μmol/L or 1. 0μmol/L) did not show any effect on intramyocytic Ca2+ concentration, and in high dose (10. 0μmol/L) presented a slight and transient inhibition. Conclusion Our data suggested that nefedine showed stronger and more sensitive effect of inhibition during insulin-induced intramyocytic calcium fluxion or during the calcium channel in the situation of active function.%目的探讨钙拮抗剂对胰岛素作用的影响及作用机理。方法用Fluo-3荧光探针负载大鼠比目鱼肌细胞,在激光扫描共聚焦显微镜下观察了细胞内Ca2+浓度的变化。结果发现钙拮抗剂硝苯地平(NF)呈剂量依赖方式抑制葡萄糖和胰岛素刺激效应。而在基础状态下,NF只有在高浓度(10.0μmol/L)时有轻微而短暂的抑制作用。结论NF对胰岛素介导的细胞内Ca2+流动或钙通道功能活跃时抑制作用更强、更敏感。
