舒血宁对糖尿病肾病大鼠肾脏保护作用的研究

         

摘要

目的 观察舒血宁(SXN)对糖尿病SD大鼠肾脏的影响并探讨其可能机制.方法 选取SPF级雄性SD大鼠腹腔注射STZ制备糖尿病模型,不同剂量SXN干预后,检测体重、血糖和24 h尿蛋白含量,Scr、BUN的变化,检测肾脏组织丙二醛(MDA)、超氧化物歧化酶(SOD)的水平.HE染色观察肾脏组织形态学改变,Western blot 检测肾脏组织中TNF-α蛋白表达.结果 大剂量SXN(12 mg/kg)治疗组(HSXN组)可明显逆转血清中Scr、BUN升高,逆转MDA的产生并升高SOD活性,减少24 h尿蛋白含量,增加体重和抑制肾脏中TNF-α的蛋白表达,而小剂量SXN(3 mg/kg)治疗组(LSXN组)仅抑制TNF-α的蛋白表达.结论 SXN对DN具有一定的保护作用,其机制可能与抑制氧化应激和TNF-α表达有关.%Objective To observe the influence of Shuxuening (SXN) on diabetic nephropathy in rats.Methods Diabetic model were established by intrap eritoneal injection of streptozotocin (STZ) in SD male rats.After treatment with different dose of SXN,Blood creatinine (Scr),urea nitrogen (BUN),superoxide dismutase (SOD) activity,malondialdehyde (MDA) level and 24 h urine protein were measured.HE staining was used to observe the morphological change of kidney.The protein expression of TNF-α were detected by Western blotting.Results High-dose SXN 12 mg/kg group (SXN H) significantly decreased the levels of serum Scr and BUN,decreased 24 h urine protein,increased the body weight,inhibited MDA content and up-regulated the SOD activity in renal tissue of diabetic rats.SXN treatment inhibited the diabetes-induced protein expression of TNF-α.Low-dose SXN 3 mg/kg group (LSXN) treatment only inhibited the diabetes-induced protein expression of TNF-α.Conclusion SXN may have protective effect on diabetic nephropathy by attenuating oxidative stress and inhibiting the protein expression of TNF-α.
