体外诱导牙髓干细胞、外胚间充质干细胞向神经分化的研究

         

摘要

目的:对牙髓干细胞(DISC)、外胚间充质干细胞(EMSC)进行体外神经诱导,分析研究二者的体外多向分化能力.方法:利用含forskolin和IBMX的诱导液体外诱导DPSC、EMSC向神经细胞分化,通过形态学观察,细胞免疫组化检测神经元标志物(NSE)、星形胶质细胞标志物(GFAP),对诱导后的细胞进行鉴定.结果:诱导后的DPSC大多数细胞均转变为类似于神经元的形态,突起伸出,而EMSC胞质收缩,胞体隆起,胞体周围伸出较长的突起.免疫组化结果显示DPSC大多数细胞NSE染色阳性,EMSC无阳性表达;GFAP染色显示DPSC少量细胞阳性,EMSC阳性.结论:DPSC、EMSC在一定的体外诱导条件下都具有向神经细胞分化的潜能,但由于迁移过程中部分性状发生改变,其诱导形成的细胞有所不同.%AIM:To investigceate the multi-direction differentiation of the dental pulp stem cell(DPSC) and ectoblast mesenchyme stem cell(EMSC)by the neural induction in intro.METHODS:DPSC and EMSC were in duced in intro by inducing fluid containing forskolin and IBMX.Then the cells were identified with the morphologic change,cell immunohistochemistry and detection of NSE of neuron marker and GFAP.RESULTS:The morphology of the induced DPSC was similar to the neuron,and their ecphyma stretched out.Cytoplasm shrinking,cyton bulging and ecphyma stretching out around cell body were found in the EMSC.NSE was positive in the DPSC and negative in the EMSC with immunohistochemistry.A few of DPSCs were found to be positive expression of GFAP.GFAP was positive in the EMSC.CONCLUSION:DPSC and EMSC have the potentiality of differentiation to nerve cell by some kind of inducing conditions in intro.The induced terminal cells are different because of the change in the immigration process of the cell character.
