维、汉两民族散发性阿尔茨海默病患者血脂水平与载脂蛋白E基因多态性关系的研究

         

摘要

目的:探讨新疆维、汉两民族阿尔茨海默病(AD)患者的血清血脂水平特点、ApoE 基因多态性对血脂代谢的影响,及两者在 AD 发病中的作用。方法对新疆地区维、汉两民族≥50岁常住居民(共8284例)进行 AD 流行病学调查,选取其中诊断为 AD 的患者157例(汉族65例、维族92例)作为 AD 组、非痴呆对照组155例(汉族68例、维族87例),应用聚合酶链反应-限制性片断长度多态性分析方法(PCR-RFLP)测定两组载脂蛋白 E(ApoE)各基因型和等位基因频率,并测定其血浆胆固醇(TC)、三酰甘油(TG)的水平。结果(1)AD 组血清 TC、TG 水平高于对照组(P <0.05);AD 组中维、汉两民族间 TC、TG 水平比较差异无统计学意义(P >0.05)。(2)ApoEε4组中 AD 患者血清 TC 水平高于对照组(P <0.05);AD 患者中 ApoEε4组、ApoEε3组与 ApoEε2组三组之间 TC 水平差异有统计学意义(F =3.65,P <0.05);在 AD 组中不同基因型维、汉族血脂水平差异均无统计学意义(P >0.05)。结论AD 患者存在血脂代谢紊乱,ApoE 基因多态性可能对血脂代谢进行着调控;ApoEε4等位基因、TC 水平升高可能是散发性 AD 的危险因素。%Objective To investigate the characteristics of serum lipid levels in patients with Alzheimer's disease (AD)of the Xinjiang Uygur and Han nationalities,and the impact of ApoE gene polymorphism on lipid me-tabolism and its role in AD pathogenesis.Methods An AD epidemiological investigation was carried out among the permanent residents(8.284 cases,age≥50 years old),including both Uygur and Han nationalities in Xinjiang.ApoE genotypes and allele frequency was detected by Polymerase chain reaction -restriction fragment length polymorphism analysis (PCR-RFLP)in potential 157 cases (65 cases Han,92cases Uygus)of AD and 155 cases(68cases Han, 87cases Uygus)of control.The concentration of serum cholesterol (TC),and triglyceride (TG)were also measured. Results (1)The levels of serum TC and TG in AD group were higher than that of control group (P <0.05),with no no statistical differences between Uygur and Han AD patients meaning(P >0.05).(2)The level of serum TC in ApoEε4 group of AD patients was higher than control group (P <0.05).There were siginificant difference among the three ApoE groups of AD patients (F =3.65,P <0.05).In the three ApoE groups,there were no differences in the levels of TC,TG among the different ethnic of Uygur and Han nationality in AD group.Conclusion AD patients are characteristized by Lipid metabolism disorder.The level of serum TC and TG in AD group is increased signficantly. ApoE gene polymorphism plays a role in the regulation of lipid metabolism,and ApoEε4 allele and higher TC level might be a risk factor in Sporadic AD.
