Purpose To establish a new technique for isolation and purification of alkamides from Echinacea Purpurea. Methods In n-hexane: ethyl acetate ;methanol: water( 2:5:5:3 ) solvent system,alkamides were isolated by high-speed counter current chromatography from petroleum ether extraction of Echinacea Purpurea root,then the compounds of alkamides purified by the prepared HPLC,and the structures were established on the basis of MS.1H NMR.and 13C NMR. Results Eight compounds were obtained by purification of 94. 43% , 96. 80% , 98. 89% , 98. 22% , 96. 42% , 98. 70% , 93. 28% and 95. 30% respectively. Conclusion HSCCC-HPLC is a very powerful technique for the isolation and purification of alkamides from Echinacea Purpurea.%目的 建立紫锥菊中多种烷基酰胺类化合物高效稳定的制备分离方法.方法 以正己烷-乙酸乙酯-甲醇-水(2∶5∶5∶3)为溶剂体系,利用高速逆流色谱对紫锥菊根石油醚萃取物进行纯化,再经制备液相分离获得各单体化合物,根据MS、1H NMR、13C NMR波谱信息确定化学结构.结果 获得纯度分别为94.43%,96.80%,98.89%,98.22%,96.42%,98.70%,93.28%和95.30%的8个烷基酰胺类化合物.结论 高速逆流色谱和制备液相色谱联用技术可高效制备分离紫锥菊中多种烷基酰胺类化合物,为紫锥菊的药理研究及质量控制奠定基础.
