目的:探讨通络保肾复方对肾小球脏层上皮细胞(POD)增殖反应及转分化生长因子-βl(TGF-β1 )mRNA表达的影响.方法:将5%含药血清培养基加入体外培养的足细胞,分别于高糖环境下脂多糖( LPS)刺激足细胞12h、24h、36h、48h后,用MTT方法检测各组的增殖情况,RT-PCR方法检测TGF-βtmRNA的表达.结果:LPS刺激12h后足细胞开始增殖,24h达高峰,48h后开始下降.通络保肾复方能够抑制足细胞增殖,以通络保肾复方大剂量组效果显著,与LPS组比较差异显著(P<0.01).同时通络保肾复方可以降低足细胞TGF-β1 mRNA的表达.本研究结果提示,通络保肾复方可以抑制高糖环境下LPS刺激诱导的足细胞增殖及降低足细胞TGF-β1 mRNA的表达,减轻足细胞的损伤,抑制细胞外基质的异常积聚和沉积,从而延缓肾小球硬化的进程.%Objective: To investigate the effect of Tongluo Baoshen Prescription on proliferation of POD and expression of TGF-B,mRNA. Methods: Preparation of serum contained 5% Tongluo Baoshen prescription was added in vitro POD. Investigate the proliferation of POD by MTT at 12、24、36、48 hours and the expression of TGF-β1 mRNAby RT-PCR. Results; The proliferation of POD started at 12 hours and reached a peak at 36 hours, declined at 48 hours. The expression of TGF-B, mRNA was inhibited. Conclusion; Tongluo Baoshen Prescription inhibite the proliferation of POD and decrease the expression of TGF-β1 mRNA delay the progression of the glomerular sclerosis.
