Objective:A metabonomics approach based on liquid chromatography coupled time-of-flight mass spectrometry (LC-TOF-MS) was utilized to obtain potential biomarkers of acute blood stasis rat and investigate metabolic mechanism of this TCM syndrome animal model.Method:SD rats were divided into normal group,blood stasis model group.Reversed-phase chromatography was used to separate the endogenous metabolites in serum.Partial least-squares to latent structure-discriminat analysis (PLS-DA) were used for data analysis.Result:Good separations were observed between the normal and model group.The major disturbed metabolic pathways were fatty acids,phosphatidylcholines and sphingolipids metabolism.Conclusion:The study provided further understanding of pathologic mechanism of the TCM syndrome animal model.%目的:通过基于液质联用技术的代谢组学方法发掘急性血瘀证大鼠动物模型的潜在生物标志物,探索该中医证候模型的发病机制.方法:SD大鼠分为正常组和模型组,应用液相色谱和质谱联用仪的代谢组学方法检测急性血瘀证大鼠血清中代谢物的变化,通过PLS-DA等模式识别方法进行数据处理.结果:与正常组比较,模型组的脂肪酸代谢、磷脂酰胆碱代谢、神经鞘磷脂代谢出现异常.结论:急性血瘀证模型中磷脂酰胆碱、脂肪酸、神经鞘磷脂等成分的代谢都受到了干扰,这些代谢途径的变化可能导致能量代谢的异常,影响血小板功能,进而导致血瘀证的发生.
