人脑胶质瘤细胞中半乳糖凝集素3和整合素αvβ3及基质金属蛋白酶2 mRNA的表达及意义

摘要

Objective To study the galactose galectin-3,integrin αvβ3 and MMP-2 mRNA expression in gliomas and their significance in the occurrence and development of gliomas. Methods 40 cases of archived formalin-fixed and paraffin-embedded specimens of glioma( grade Ⅰ12 cases,gradeⅡ11 cases,gradeⅢ13 cases and gradeⅣ4 cases)and 5 cases of normal brain tissues of patients with brain tauma were obtained from decompression in Kailuan of Hebei United University Gen-eral Hospital from 2012 to 2013. ISHH was used to detect the galectin-3,integrinαvβ3 and MMP-2 mRNA expression in glio-ma and normal brain tissues,and the relationship among the three was analyzed. Results The galectin-3,integrin αvβ3 and MMP-2 mRNA expression were negative in normal brain tissues,but were positive in glioma specimens. The LI of galectin-3, integrin αvβ3 and MMP-2 mRNA expressions in gradeⅢand gradeⅣwere higher than those of gradeⅠand gradeⅡ〔(27. 3±22. 7)% vs. (4. 8±3. 2)%,(29. 2±21. 1)% vs. (5. 4±3. 8)%,(29. 9±24. 0)% vs. (6. 4±5. 3)%,P<0. 05〕. The linear correlation analysis showed that galectin-3 mRNA expression was positively correlated with integrin αvβ3 and MMP-2 mRNA expression(r=0. 718 and 0. 790,P<0. 05). Integrin αvβ3 mRNA expression was positively correlated with MMP-2 mRNA expression(r=0. 812,P <0. 05). Conclusion Galectin-3,integrin αvβ3 and MMP-2 mRNA expression are positively correlated with the malignancy level of glioma. Galectin-3,integrin αvβ3 and MMP-2 mRNA expression,with synergistic effect,can together promote the development,infiltration and angiogenesis and glioma,which is conducive to the evaluation and judgment of the malignant behavior of glioma.%目的:探讨半乳糖凝集素3(galectin-3)、整合素αvβ3及基质金属蛋白酶2(MMP-2)mRNA在人脑胶质瘤细胞中的表达及三者在胶质瘤发生发展中的作用。方法选取2012-2013年河北联合大学附属开滦总医院病理科存档的经甲醛固定、石蜡包埋的胶质瘤标本40例(Ⅰ级12例,Ⅱ级11例,Ⅲ级13例,Ⅳ级4例)及神经外科收治的脑创伤患者内减压术中得到的5例正常脑组织。采用原位杂交组织化学技术( ISHH)检测胶质瘤组织和正常脑组织中galectin-3、整合素αvβ3、MMP-2 mRNA的表达情况,并分析三者之间的关系。结果正常脑组织中galectin-3、整合素αvβ3、MMP-2 mRNA的表达均为阴性;而胶质瘤标本中galectin-3、整合素αvβ3、MMP-2 mRNA的表达均为阳性。Ⅲ~Ⅳ级胶质瘤标本中galectin-3、整合素αvβ3、MMP-2 mRNA表达的阳性指数( LI)均高于Ⅰ~Ⅱ级胶质瘤标本〔(27.3依22.7)%与(4.8依3.2)%,(29.2依21.1)%与(5.4依3.8)%,(29.9依24.0)%与(6.4依5.3)%,P<0.05〕。直线相关分析显示,galectin-3 mRNA的表达与整合素αvβ3、MMP-2 mRNA的表达均呈正相关( r值分别为0.718和0.790,P<0.05);整合素αvβ3 mRNA的表达与MMP-2 mRNA的表达呈正相关(r=0.812,P<0.05)。结论胶质瘤恶性程度越高,galectin-3、整合素αvβ3、MMP-2 mRNA表达量越多,均从分子水平共同促进胶质瘤的发展、浸润及血管生成,三者之间起协同作用,有利于从分子水平对人脑胶质瘤的恶性生物学行为进行评价和判断。
