为考察杠柳苷元经大鼠肝微粒体体外代谢的特点,对其代谢产物进行了定性研究.建立肝微粒体温孵体系,采用超高效液相色谱和四级杆飞行时间串联质谱(ultra performance liquid chromatography/quadrupole-time-of-flight mass spectrometry,UPLC-Q-TOF/MS)技术,结合保留时间、精确质量数和离子的碎片特征对孵育后的代谢产物进行结构鉴定.结果表明:杠柳苷元经肝微粒体温孵后,可以检测到除原型以外的4个I相代谢产物;杠柳苷元经大鼠肝微粒体温孵后,主要的代谢途径为脱水、脱氢和甲基化.本研究为其体内代谢的研究提供了基础.%The in vitro metabolites of periplogenin in rat liver microsomes were investigated, and qualitative study of the metabolites were carried out.After establishing the rat liver microsomal incubation system, the metabolites of periplogenin in rat liver microsomes were detected by ultra performance liquid chromatography/quadrupole-time-of-flight mass spectrometry (UPLC-Q-TOF/MS) combined with retention time, exact mass number and fragmentation characteristics of the ions.The results ithat besides the parent component, four metabolites were determined as phase I metabolites in the rat liver microsomal incubation.The dehydration, dehydrogenation and methylation were the major phase I metabolic pathways of periplogenin in vitro.The present study provided the theoretical basis for metabolism in vivo.
