四季草颗粒纯化工艺改进研究

         

摘要

目的 探讨新纯化工艺替代原纯化工艺用于四季草颗粒生产工艺的可行性.方法 以槲皮苷和总固体为评价指标,对5种常用澄清剂进行比较试验.采用正交试验设计对最优澄清剂进行条件优化,优选出最佳澄清剂.以最优澄清工艺与传统的醇沉工艺进行药效学试验比较,对吸附澄清工艺进行综合评价.结果 吸附澄清剂为壳聚糖,最佳工艺条件为提取液浓缩至1:8[生药(g):药液(mL)],提取液pH为4,每1 L药液加入1%壳聚糖冰醋酸溶液100 mL.结论 壳聚糖吸附澄清工艺具有工艺简单、成本低、有效成分损失少的特点,可代替传统工艺用于四季草颗粒的制备.%Objective To investigate a new purification process to substitute the original process for Sijicao Granules. Methods With quercetin and total solids as the evaluation indexes, the best clarifier was selected from five kinds of commonly used clarifier. The optimal clarifier was optimized by orthogonal test. Finally, the best clarification process was compared with the traditional alcohol precipitation process, and the comprehensive evaluation of the adsorption clarification process was carried out. Results The optimized adsorption clarifier was chitosan, and the conditions were as follows: the extract was concentrated to 1 : 8[crude drug(g) : liquid(mL) ], the pH of extraction solution was 4, and 100 mL 1% chitosan glacial acetic acid solution was added into every 1 L drug solution. Conclusion The chitosan adsorption clarification process has the characteristics of simple process, low cost, and less loss of active ingredient, which can be used in the preparation of Sijicao Granules instead of the traditional process.
