青蒿琥酯对糖尿病大鼠视网膜细胞凋亡的作用

摘要

目的 观察青蒿琥酯对糖尿病大鼠视网膜细胞凋亡的作用.方法 将18只健康雄性Sprague Dawley大鼠完全随机分为空白对照组、模型对照组及药物治疗组,各6只.模型对照组和药物治疗组腹腔注射链脲佐菌素建立糖尿病大鼠模型,3个月后,药物治疗组大鼠腹腔注射青蒿琥酯20 mg/(kg·d),正常对照组和模型对照组大鼠腹腔注射等体积0.9%氯化钠注射液,10 d后摘除眼球,过碘酸Schiff(PAS)染色观察视网膜细胞形态学的变化,实时荧光定量聚合酶链式反应(qRT-PCR)方法检测视网膜组织中B淋巴细胞瘤-2蛋白(Bcl-2)和Bcl-2相关x蛋白(Bax)mRNA水平.结果 PAS染色显示,模型对照组视网膜发生病理改变,药物治疗组较模型对照组视网膜病变有所改善.模型对照组大鼠视网膜组织中Bcl-2mRNA水平明显低于空白对照组大鼠[(0.72±0.05)比(1.23±0.43)],药物治疗组大鼠Bcl-2 mRNA水平(2.46 ±0.34)明显高于模型对照组和空白对照组大鼠,差异有统计学意义(P<0.05或P<0.01);药物治疗组大鼠视网膜组织中Bax mRNA水平明显低于模型对照组和空白对照组大鼠[(0.51±0.16)比(1.13±0.26)、(0.98±0.07)],差异有统计学意义(P<0.01),而模型对照组和空白对照组间差异无统计学意义(P>0.05).结论 青蒿琥酯对糖尿病大鼠视网膜细胞凋亡具有一定的抑制作用.%Objective To investigate the effects of artesunate (ART) on apoptosis of retinal cells in diabetes rats.Methods Totally 18 healthy male Sprague Dawley rats were randomly divided into control group,model group and ART treated group.The rats in model group and ART treated group were given intraperitoneal injection of streptozotocin to establish diabetes model;3 months later,the rats in ART treated group were given intraperitoneal injection of ART [20 mg/(kg · d)],and the rats in control group and model group were given intraperitoneal injection of 0.9% sodium chloride injection.Ten days later,the eyeballs were isolated,periodic acidSchiff (PAS) stain was used to observe the morphological changes of retina,quantitative reverse transcription polymerase chain reaction (qRT-PCR) was used to detect the mRNA levels of B-cell lymphoma 2 (Bcl-2) and Bcl-2-associated x (Bax).Results Pathologically changes of retinal morphology was observed in model group in PAS stain,which was significantly improved in ART treated group.In model group,the Bcl-2 mRNA level in retinal tissue was significantly lower than that in control group [(0.72 ± 0.05) vs (1.23 ± 0.43)],in ART treated group,it was significantly higher (2.46 ± 0.34) than that in model group and control group (P < 0.05 or P <0.01);the Bax mRNA level in retinal tissue in ART treated group was significantly lower than that in model group and control group [(0.51 ±0.16) vs (1.13-±0.26),(0.98-±0.07)] (P <0.01),while was not significantly different between model group and control group (P > 0.05).Conclusion Artesunate has inhibitory function on apoptosis of retinal cells in diabetes rats.
