丁酸钠对重症急性胰腺炎大鼠胰腺腺泡细胞凋亡的影响

         

摘要

目的 探讨丁酸钠(NaB)对重症急性胰腺炎(SAP)大鼠胰腺腺泡细胞凋亡的影响.方法 30只SD大鼠分成对照组、SAP组及NaB治疗组,每组各10只,采用牛磺胆酸钠逆行胰胆管注射制作SAP动物模型,NaB治疗组为SAP制模5 min后通过股静脉注射NaB(5 mg/kg).检测血清淀粉酶、胰腺湿干重比值,利用Kusske方法对胰腺组织的病理学改变:水肿、出血、坏死、炎细胞浸润进行计分,TUNEL法检测胰腺腺泡细胞凋亡.结果 SAP组血清淀粉酶与对照组比较明显升高,差异有高度统计学意义(P < 0.01),NaB治疗组血清淀粉酶较SAP组明显下降,差异有统计学意义(P < 0.05);SAP组胰腺组织湿/干重比值较对照组下降,差异有高度统计学意义(P < 0.01),NaB治疗组胰腺组织湿/干重比值较SAP组明显下降,差异有统计学意义(P < 0.05).SAP组胰腺损伤积分明显高于对照组,差异有高度统计学意义(P < 0.01),NaB治疗组胰腺损伤积分明显低于SAP组,差异有统计学意义(P < 0.05).SAP组胰腺腺泡细胞凋亡指数高于对照组,差异有高度统计学意义(P < 0.01),NaB治疗组胰腺腺泡细胞凋亡指数显著高于SAP组,差异有统计学意义(P < 0.05).结论 NaB可以促进SAP大鼠胰腺腺泡细胞凋亡,减轻中性粒细胞活化,降低SAP严重程度.%Objective To investigate the effects of sodium butyrate on apoptosis of pancreatic cells in rats with severe acute pancreatitis (SAP). Methods 30 SD rats were randomly divided into the control group, SAP model group and NaB treatment group, with 10 cases in each group. The SAP models rats were established by retrograde injection of sodium tauro-cholate into biliopancreatic duct respectively, sodium butyrate was given to the rats in SAP model group by femoral vein injection 5 minutes after the establishment of SAP model. The levels of serum amylase and the wet/dry weight ratio of pancreas were detected. Pancreatic histopathological scores including edema, hemorrhage, necrosis, inflammatory were evaluated by Kusske method. Apoptosis of pancreatic cells was detected by TUNEL method. Results The serum amylase in SAP group was higher than that in control group, the difference was statistically significant (P< 0.01), the serum amylase in NaB treatment group was lower than that in SAP group, the difference was statistically significant (P < 0.05); the wet/dry weight ratio in SAP group was higher than that in control group, the difference was statistically significant (P < 0.01), the wet/dry weight ratio in NaB treatment group was lower than that in SAP group, the difference was statistically significant (P < 0.05); pancreatic injury integral in SAP group was higher than that in control group, the difference was statistically significant (P < 0.01), pancreatic injury integral in NaB treatment group was lower than that in SAP group, the difference was statistically significant (P < 0.05); the apoptotic index in SAP group was higher than that in control group, the difference was statistically significant (P< 0.01), the apoptotic index in NaB treatment group was lower than that in SAP group, the difference was statistically significant (P < 0.05). Conclusion NaB can contribute to the apoptosis of rat pancreas acinar cells, alleviate the activation of neutrophil and relieve the severity of SAP.
