丙型肝炎病毒高变区1模拟表位7肽对LPS刺激RAW264.7细胞炎症因子影响的研究

         

摘要

目的 观察丙型肝炎病毒(HCV)高变区1(HVR1)模拟表位7肽(7P)对内毒素/脂多糖(LPS)诱导的小鼠单核/巨噬细胞株(RAW264.7)细胞释放细胞因子的影响.方法 体外培养小鼠RAW264.7细胞,用LPS刺激小鼠RAW264.7细胞分泌TNF-α、IL-6,用不同浓度(5.0、10.0、20.0、40.0、80.0、160.0 μg/mL)的7P进行干预,应用单溶液细胞增殖(MTS)法检测7P对细胞活力的影响,应用酶联免疫吸附实验(ELISA) 法检测细胞培养上清中促炎细胞因子的含量.结果 7P可抑制LPS刺激小鼠RAW264.7细胞释放TNF-α和IL- 6,并呈一定的量效关系,其影响细胞因子释放作用与细胞毒作用无关.结论 7P呈浓度依赖性地抑制LPS刺激小鼠RAW264.7细胞释放TNF-α和IL-6.%Objective To observe the effect of synthetic heptapeptide (7P) from HCV HVR1 on inflammatory cytokines release from RAW264.7 cells induced by lipopolysaccharides (LPS). Methods Murine RAW264.7 cell lines were cultivated and released cytokines TNF-α and IL-6 induced by LPS. The effect of 7P on which were detected by enzyme linked immunosolbent assay (ELISA). Results 7P could inhibit the content of TNF-α and IL-α in adose-dependent manner markedly. The effects of 7P on inflammatory cytokines released from RAW264.7 cells had nothing to do with the cytotoxic action. Conclusion 7P can inhibit TNF-α and IL-6 release induced by LPS in a dose-dependent manner.
