FAK在良性和恶性黑素细胞肿瘤中的表达

目的:检测FAK在良性和恶性黑素细胞肿瘤中的表达.方法:用RT- PCR和免疫组化的方法检测25例恶性黑素瘤和25例痣细胞痣和9例发育不良痣中局部黏附斑激酶(FAK)mR州A和蛋白的表达.结果:恶性黑素瘤组织中FAK mRNA和蛋白均表达增高,与痣细胞痣和发育不良痣有明显差异,而后两者间表达水平无明显差异.结论:FAK表达增高发生于肿瘤的早期,可作为细胞恶性转化的标志,发育不良痣不代表一种癌前病变状态.%Objective: To detect the mRNA and protein level of focal adhesion kinase (FAK) in benign and malignant melanoma. Methods: RT- PCR and unmunohistochemistry technique were used to detect the expression of FAK mRNA and protein in 25 cases of malignant melanoma, 23 cases of melanocytic nevus and 9 cases of dysplastic nevus. Results: FAK mRNA and protein levels were significantly increased, in malignant melanoma, compared with benign nevus and dysplastic nevus. There was no significant difference between benign nevus and dysplastic nevus. Conclusion: The over- expression of FAK in early melanoma can be used as a mark of cell malignant transformation. Dysplastic nevus is not premalignant.
