以铁皮石斛花为材料,通过单因素和正交试验对超声-乙醇法提取铁皮石斛花总黄酮的工艺进行研究.结果表明,超声波乙醇法提取铁皮石斛花总黄酮的最佳工艺为乙醇体积分数80%,料液比1∶35(g∶mL),超声温度40 ℃,超声时间50 min,在此条件下石斛花总黄酮提取率最高,为2.31%.对超声-乙醇法提取的铁皮石斛花总黄酮进行抗氧化的活性测定,试验结果显示铁皮石斛花总黄酮对DPPH·、·OH有明显的清除作用,当质量浓度达10 mg/mL时,其清除率分别为89.77%和80.01%.%UsingDendrobium officinale flowers as raw materials, the extraction process of total flavonoids from D. officinale flowers with ultrasonicethanol synergistic extraction was studied by single factor and orthogonal experiments. The results showed that the optimum process parameters were ethanol concentration 80%, solid-liquid ratio 1∶35 (g∶ml), ultrasonic temperature 40 ℃, time 50 min. Under the above conditions, the extraction rate of total flavonoids from D. officinale flowers was the highest of 2.31%. The antioxidant activity of the total flavonoids from D. officinale flowers were studied, and the total flavonoids had better scavenging ability on DPPH·and·OH. When the concentration was 10 mg/ml, the scavenging rate of DPPH·and·OH were 89.77% and 80.01%, respectively.
