富勒烯衍生物纳米颗粒水悬液对细菌生长的抑制作用

     

摘要

测定了二加成亚甲基富勒烯[60]二膦酸四乙酯(Bis-methanophosphonate fullerene,BMPF)的纳米水悬液(nano-BMPF)和富勒醇纳米水悬液(nano-Fullerol)对细菌生长的影响.结果表明,nano-BMPF和nano-Fullerol在黑暗条件下可抑制革兰氏阳性菌金黄色葡萄球菌的生长,且呈现浓度梯度依赖性,半抑制浓度IC50值分别为9.1和4.2 μmol/L; nano-Fullerol对金黄色葡萄球菌生长的抑制可能与活性氧无关,nano-BMPF对金黄色葡萄球菌生长的抑制则可能与超氧阴离子自由基(O2·-)有关.在黑暗条件下,二者对革兰氏阴性菌大肠杆菌的生长无影响.研究结果表明,这两种富勒烯衍生物纳米颗粒作为抗生素在生物医用领域具有潜在的应用前景.%In recent years, the biological activities of fullerene nanoparticles have received extensive attention, but there are few reports on aqueous nanoparticle suspensions of [60] fullerene derivatives and bacterial growth. The antibacterial effects of two aqueous nanoparticle suspensions of [60] fullerene derivatives, bis-methanophosphonate fullerene(BMPF) and fullerol, were studied for the first time by the bacterial growth inhibition test. The results show that both nano-BMPF and nano-fullerol could dose-dependently inhibit the growth of the gram-positive bacteria S. aureus under dark condition. The 50% inhibition concentrations of these two nanoparticle suspensions were 9.1 and 4.2 μmol/L, respectively. In order to explore their antibacterial mechanism, three different scavengers of reactive oxygen species were tested, including singlet oxygen inhibitor azide, hydroxyl free radical inhibitor mannitol and superoxide anion inhibitor superoxide dismutase.It was revealed that the antibacterial activity of nano-fullerol probably did not involve reactive oxygen species,but superoxide anion might be involved in the antibacterial action of nano-BMPF. In addition, both aqueous nanoparticle suspensions couldn't inhibit the growth of the gram-negative bacteria E. coli under dark condition. These results indicate that nanoparticles of different fullerene derivatives prevented bacterial growth via distinct mechanisms, and they might be potentially used in the biomedical field as novel antibacterial reagents.
