Objective To investigate the role of PTEN in aldosterone ( Ald)-induced proliferation of cardiac fibroblasts(CFs) and to explore the effects of spironolactone (Spi) on the expression of PTEN.Methods The effects of Ald and Spi on proliferation were measured by MTT colorimetric assay, synthesis of collagen was observed by the hydroxyproline concentration, PTEN mRNA and protein were tested by RT-PCR and Western blot respectively.Results Compared with control group, the proliferation of the fibroblasts and the hydroxyproline concentration were significantly increased, while PTEN mRNA and protein were down-regulated in Ald group (P <0.05 ).Compared with Ald group, the proliferation of the fibroblasts and hydroxyproline concentration were significantly inhibited,while PTEN mRNA and protein were increased in Ald + Spi group ( P < 0.05 ).Conclusion Exogenous aldosterone stimulated cardiac fibroblasts proliferation、 collagen synthesis and inhibited the expression of PTEN mRNA、protein.The effects of Ald on PTEN might be mediated through genomic way.%目的 研究醛固酮(Ald)诱导的心肌成纤维细胞(CFs)增殖中PTEN mRNA、蛋白表达及螺内酯(Spi)对其表达的影响,从而探讨Ald对心肌成纤维细胞PTEN表达的影响.方法 用体外培养新生SD大鼠的CFs,以Ald诱导其增殖和胶原合成,并加入Spi干预,用RT-PCR和Western Blot分别测定PTEN mRNA和蛋白表达.结果 与对照组相比,Ald组的CFs增殖和羟脯氨酸含量明显增加(P<0.05),并旱浓度依赖性降低PTEN mRNA和蛋白表达(P<0.05);与Ald组相比,Ald+Spi组的CFs增殖和羟脯氨酸含量明显减少(P<0.05),同时其PTEN mRNA、蛋白表达均明显升高(P<0.05).结论 醛固酮可通过盐皮质激素受体(MR)介导的基因组通路降低PTEN表达发挥其致心肌纤维化作用.
