GSTT1基因型和吸烟状况与肺癌易感性关系

         

摘要

Objective To investigate the relationship between the genetic polymorphism of GSTT1 and the susceptibility to lung cancer and potential synergetic interactions. Methods PCR-RFLP were used to identify GSTT1 genotype in 100 cases of lung cancer and 135 controls. Results The frequency of GSTT1 ( - ) genotype in lung cancer group and controls was respectively 57% and 41.5% ( P < 0. 05 ) . In people with GSTT1 ( - ) genotype, smoking markedly increased the risk of lung cancer ( P < 0. 05 ) . In smokers, GSTT1 ( - ) genotype significantly increased the risk of lung cancer( P < 0. 01 ) . In smokers > 20 packyears, GSTT1 ( - ) genotype remarkably increased the risk of lung cancer (P < 0. 01 ) . Conclusions GSTT1 ( - ) genotype increases the susceptibility to lung cancer and there may be a synergetic interaction between smoking and GSTT1 ( - ) genotype.%目的 探讨GSTT1基因多态性与肺癌遗传易感性的关系及GSTT1基因多态性和吸烟在肺癌易感性中的交互作用.方法 采用病例对照和PCR-RFLP方法对100例肺癌患者和135例健康对照者外周血的GSTT1基因型进行检测,并评价其与吸烟和肺癌遗传易感性的关系.结果 GSTT1(-)基因型在肺癌组为57%,显著高于对照组的41.5%(P<0.05).GSTT1(-)基因型的吸烟者较不吸烟者患肺癌的风险增加3.64倍(P<0.05).吸烟者携带GSTT1(-)基因型较携带GSTT1(+)基因型患肺癌的风险增加3.28倍(P<0.01).吸烟量≥20包/年的人群中携带GSTT1(-)基因型者较携带GSTT1(+)基因型者患肺癌的风险增加7.81倍(P<0.01).结论 GSTT1(-)基因型增加肺癌遗传易感性,吸烟与GSTT1(-)基因型间存在交互作用,二者协同增加患肺癌的风险性.
