PTPIP51、p-Raf-1及ERK1/2在口腔鳞状细胞癌中的表达

         

摘要

Objective To study the expression of PTPIP51 , p-Raf-1 and ERK1/2 in oral squamous cell carcinoma(OSCC)and their impact on invasion and migration of OSCC. Methods The expression of PTPIP51 and p-Raf-1 was detected by immunofluorescence histochemistry and confocal laser scanning microscope in 32 tumor specimens and their adjacent normal mucosa specimens. The PTPIP51-positive cells of the carcinoma and the surrounding stroma were identified on the basis of specific morphological feature by means of HF staining and immunohistochemistry(SP method). The total RNA was extracted from these specimens,and the expression of PTPIP51 mRNA and ERK1/2 mRNA was detected by real-time PCR. Results PTPIP51 and p-Raf-1 proteins were coexpressed in malignant proliferating cells,immune cells and surrounding endothelial cells of vessels in OSCC. The expression levels of PTPIP51 mRNA and ERK1/2 mRNA in tumor specimens were significantly higher than those of the adjacent normal mucosa specimens. There was a positive correlation between the expression of PTPIP51 mRNA and ERK1/2 mRNA(r=0. 641,P<0. 05). The expression levels of PTPIP51 mRNA and FRK1/2 mRNA in OSCC were higher in group with lymph node metastasis than that in group without lymph node metastasis ( P<0. 05). The expression of PTPIP51and ERK1/2 mRNA was correlated with TNM staging and the lymph node metastasis,but not with age,gender and the pathological grading. Conclusion PTPIP51 and Raf-1 may play a key role in the pathogenesis and development of OSCC via Ras/Raf/MEK/FRK pathway(extracellullar signai-regulated kinase pathway) , and they are closely related to invasion and metastasis of Oscc.%目的 探讨PTPIP51和p-Raf-1及ERK1/2在口腔鳞状细胞癌中的表达及对口腔鳞癌侵袭、转移的影响.方法 收集32例口腔鳞状细胞癌及其配对癌旁正常口腔黏膜组织.用免疫荧光组织化学染色激光共聚焦显微镜检测PTPIP51与p-Raf-1蛋白在组织切片内表达及定位;PBS冲洗后对标本进行苏木精-伊红染色,普通光镜对比观察组织结构及细胞形态,同时用免疫组织化学方法(SP法)检测PTPIP51蛋白在组织和细胞内的定位;Real-time PCR检测PTPIP51与ERK1/2基因mRNA在肿瘤组织及癌旁正常口腔黏膜组织的表达情况.结果 PTPIP51与p-Raf-1蛋白在癌巢周围增殖旺盛的肿瘤细胞、间质免疫细胞及部分脉管壁内皮细胞表达阳性且2种蛋白在细胞中共定位表达.同时发现PTPIP51与ERK1/2基因 mRNA在口腔鳞癌组织表达高于配对癌旁正常口腔黏膜组织,差异具有统计学意义(P<0.05);ERK1/2及PTPIP51基因mRNA表达呈正相关(r=0.641,P<0.05);两者的表达水平与患者的性别、年龄,病理分级无关,与肿瘤的TNM分级及有无淋巴结转移有关(P<0.05).结论 PTPIP51与Raf-1可能通过Ras/Raf/MEK/ERK信号通路参与口腔鳞状细胞癌的发生、发展过程,且2种蛋白与肿瘤的侵袭、转移有密切关系.
